摘 要:目的 研究貝健膠囊對小鼠抽動障礙(TD)模型的治療作用,通過檢測模型小鼠腦內(nèi)神經(jīng)遞質(zhì)多巴胺(DA)的量,探討貝健膠囊治療TD的作用機(jī)制。方法 咖啡因誘導(dǎo)小鼠TD模型,采用自主行為實(shí)驗(yàn)和攀爬實(shí)驗(yàn)研究貝健膠囊對模型小鼠的影響;采用高效液相色譜法測模型小鼠腦組織中DA的量。結(jié)果 貝健膠囊能明顯抑制咖啡因引起的小鼠攀爬行為(P<0.05),并顯著減少自發(fā)活動次數(shù)(P<0.05);降低模型小鼠腦內(nèi)DA的量(P<0.05)。結(jié)論 貝健膠囊對咖啡因誘導(dǎo)的小鼠TD模型具有顯著的治療作用,降低腦內(nèi)多巴胺的量可能是其治療TD的作用機(jī)制。

Abstract: Objective To observe the therapeutic effects of Beijian Capsule on mouse with tic disorders(TD),and to explore the mechanism of Beijian Capsule for treatment of TD by determining the neurotransmitter dopamine content in mouse brain.Methods TD model of mouse was established by gastric perfusion of caffeine.The climbing test and spontaneous behavior test were performed to study the effects of Beijian Capsule on the mouse model.Dopamine in mouse brain was determined by high performance liquid chromatography- fluorescence detection(HPLC-FLD).Results Beijian Capsule significantly inhibited the mice abnormal climbing behavior caused by caffeine(P<0.05),and markedly reduced the spontaneous movements (P<0.05).Beijian Capsule also decreased the dopamine content in mouse brain(P<0.05).Conclusion Beijian Capsule has significant therapeutic effects on mouse with tic disorders, and the mechanism may be related to its decreasing dopamine content in mouse brain.
