摘 要:目的 首次以藁本內(nèi)酯純度和轉(zhuǎn)移率為主要考察指標(biāo),探討應(yīng)用超臨界 CO2 萃取法從當(dāng)歸中提取總苯酞的工藝。方法 采用單因素和 L9(3)4 正交設(shè)計(jì)進(jìn)行試驗(yàn),考察藥材粒度、萃取壓力、萃取時(shí)間、萃取溫度等因素對(duì)藁本內(nèi)酯純度和轉(zhuǎn)移率的影響,優(yōu)化最佳提取工藝。采用高效液相色譜法分析,色譜條件為 Accurasil C18 柱(250 mm×4.6 mm,5 μm),流動(dòng)相為甲醇-0.05%磷酸水溶液,柱溫30 ℃,檢測(cè)波長320 nm,體積流量1.0 mL/min。結(jié)果 超臨界 CO2 萃取法的最佳工藝條件為:萃取壓力10 MPa,萃取溫度50 ℃,萃取時(shí)間20 min。結(jié)論 超臨界 CO2 流體工藝萃取的當(dāng)歸總苯酞純度和轉(zhuǎn)移率可達(dá)80%以上。

[Abstract]
Abstract: Objective To establish the optimal supercritical CO2 extraction of total phthalide from Angelica sinensis.The ligustilide purity and transfer rate in general phthalide were used as the evaluation indexes.Methods The optimum extraction conditions,such as sizes of crush materials,the extraction pressure,extraction temperature,and extraction time were investigated by L9(3)4 orthogonal test.The HPLC was performed on a Accurasil C18 column(250 mm×4.6 mm,5 μm).The mobile phase consisted of methanol-0.05% phosphoric acid water.The column temperature was 30 ℃.Examination wavelength was 320 nm.The flow rate was 1.0 mL/min. Results The optimal supercritical CO2 extraction conditions were: extraction pressure,10 MPa;extraction temperature,50℃;extraction time,20 min.Conclusion It is feasible that supercritical CO2 extraction could be used in the extraction of total phthalide from A.sinensis,its purity and transfer rate are over 80%.
