[摘要]
目的 確定川赤芍、赤芍、白芍抑制脂多糖(LPS)誘導(dǎo)大鼠肺泡巨噬細(xì)胞NR8383過(guò)度激活的“藥效成分組”。方法 以川赤芍、赤芍、白芍不同提取物L(fēng)C-MS圖譜峰面積及其抑制LPS誘導(dǎo)的NR8383細(xì)胞活力增強(qiáng)為基礎(chǔ),采用偏最小二乘回歸法,對(duì)LC-MS圖譜和抑制細(xì)胞增殖的內(nèi)在聯(lián)系進(jìn)行譜效分析。結(jié)果 LC-MS圖上的X11、X6、X1、X2、X9、X3這6個(gè)成分組成的“藥效成分組”對(duì)川赤芍、赤芍、白芍抑制NR8383細(xì)胞增殖藥效的貢獻(xiàn)較大。結(jié)論 “藥效成分組”的研究顯示,同基源的赤芍、白芍在抑制LPS誘導(dǎo)的NR8383細(xì)胞增殖方面具有相同的化學(xué)成分(X1、X2、X9、X3),但同時(shí)也有某些成分(X11、X6)僅能在赤芍中檢測(cè)到,這提示不同的化學(xué)成分及含量差異可能是造成赤芍、白芍功效不同的物質(zhì)基礎(chǔ)。

[Abstract]
Objective To study the “effective components group”of Paeoniae Radix Rubra, Paeoniae Radix Veitchii, and Paeoniae Radix Alba in the inhibiting proliferation of NR8383, which was induced by lipopolysaccharide (LPS). Methods The inherent relationship between LC-MS fingerprint and proliferation inhibition with partial least squares regression were analysed, which were based on LC-MS fingerprint peak areas of all samples and the inhibiting of cell activity increase of NR8383 of Paeoniae Radix Rubra, Paeoniae Radix Veitchii, and Paeoniae Radix Alba. Results Effective components group consists of six peaks X11, X6, X1, X2, X9, and X3 of the fingerprint, which played the most inhabitation of NR8383 proliferation. Conclusion “Effective components group” indicates that Paeoniae Radix Rubra and Paeoniae Radix Alba with the same origin contain the common ingredients (X1, X2, X9, and X3) in the inhibiting proliferation of NR8383. However, X11 and X16 only can be detected in Paeoniae Radix Rubra, which can be considered as the different material basis in efficacy between Paeoniae Radix Rubra and Paeoniae Radix Alba.
