[摘要]
類風(fēng)濕性關(guān)節(jié)炎(rheumatoid arthritis,RA)、節(jié)段性腸炎(Crohn′s disease,CD)等疾病具有類似的發(fā)病機(jī)制。腫瘤壞死因子(tumor necrosis factor,TNF)在這些疾病的發(fā)生發(fā)展中具有非常重要的作用。TNF分子在細(xì)胞因子免疫調(diào)節(jié)網(wǎng)絡(luò)中位于中樞環(huán)節(jié),是重要的炎癥介質(zhì),過量的TNF導(dǎo)致炎癥的發(fā)生。目前上市的TNF抑制劑種類繁多,治療效果顯著。然而隨著TNF抑制劑廣泛應(yīng)用,這類藥物的安全性逐漸受到重視。本文綜述了TNF與疾病關(guān)系、上市的TNF蛋白抑制劑分子結(jié)構(gòu)、臨床應(yīng)用等方面,重點(diǎn)關(guān)注藥物潛在的不良反應(yīng),并對(duì)近期新型TNF抑制劑的研究開發(fā)進(jìn)行總結(jié)。

[Abstract]
The pathogenesis mechanisms of diseases like rheumatoid arthritis (RA), Crohn’s disease (CD) are quite similar. Tumor necrosis factor (TNF) is likely to be of fundamental importance on the generation and development and plays a key role on the cytokine immune network. High level of TNF could induce inflammation. There are some TNF inhibitors marketed or in research, and both of them had remarkable efficacy. Along with TNF inhibitors being widely used, it’s more important to concern about the safety profiles. Therefore, this article reviews the relationship between TNF and diseases, and five TNF protein inhibitors available through structures and clinical use, keeping eyes on their possible side effect and providing some new TNF inhibitors with bright future.
