[摘要]
目的 探討英夫利西單抗對哮喘模型豚鼠抗腫瘤壞死因子-α(TNF-α)的影響。方法 豚鼠50只,雌雄各半,隨機(jī)分為5組:對照組,模型組,英夫利西單抗低、中、高劑量(5、10、15 mg/kg)組,每組10只。模型組和英夫利西單抗各劑量組ip給予10%卵蛋白以致敏,對照組給予等量生理鹽水;模型組和英夫利西單抗各劑量組再分兩次給予2%卵蛋白溶液霧化吸入,對照組以等量生理鹽水替代。每次激發(fā)前英夫利西單抗各劑量組ip相應(yīng)劑量的英夫利西單抗,第二次激發(fā)后24 h處死動物,對比各組血清、組織勻漿和肺泡灌洗液中TNF-α水平。結(jié)果 模型組血清、組織勻漿、肺泡灌洗液中TNF-α濃度高于對照組(P<0.05),而英夫利西單抗各劑量組血清、組織勻漿、肺泡灌洗液中TNF-α的濃度低于模型組(P<0.05)。結(jié)論 TNF-α與豚鼠哮喘形成密切相關(guān);使用英夫利西單抗可以減少豚鼠哮喘模型血清、組織勻漿、肺泡灌洗液中TNF-α的產(chǎn)生。

[Abstract]
Objective To investigate the effect of Infliximab on TNF-α of guinea pigs asthma model. Methods Guinea pigs (50), half male and female, were randomly divided into five groups, control, model, low-, mid-, and high-dose Infliximab (5, 10, and 15 mg/kg) groups (n=10). Model and the Infliximab groups were ip administered with 10% ovalbumin to sensitivate, and the control group was given physiological saline. Model and Infliximab groups were atomizing inhalated twice with 2% ovalbumin solution, and the control group was given the same dose physiological saline. Infliximab groups were ip administered with appropriate doses of Infliximab before excitation each time, and animals were killed 24 h after the second excitation. TNF-α was detected in serum, tissue, and bronchoalveolar lavage fluid. Results TNF-α concentration in the serum, tissue homogenates, and bronchoalveolar lavage fluid in the model group was higher than that in the control group (P < 0.05). However, TNF-α concentration in the serum, tissue homogenates, and bronchoalveolar lavage fluid of the Infliximab group was lower than that in the model group (P <0.05). Conclusion TNF-α is related to the formation of the guinea pig asthma, and Infliximab could reduce TNF-α of serum, tissue homogenates, and bronchoalveolar lavage fluid in guinea pig model of asthma.
