[摘要]
目的 建立山楂中化學(xué)成分的超高液相/電噴霧–飛行時(shí)間/質(zhì)譜(UPLC/ ESI-TOF/MS)的分析方法。方法 Acquity UPLC BEH C18色譜柱(100 mm×2.1 mm,1.7 μm),流動(dòng)相0.1%甲酸水溶液–乙腈,梯度洗脫;體積流量0.3 mL/min,柱溫50 ℃,檢測(cè)波長(zhǎng)范圍260~360 nm,進(jìn)樣量1 μL。ESI離子源,飛行時(shí)間質(zhì)譜檢測(cè)器,對(duì)比對(duì)照品進(jìn)行鑒別。結(jié)果 共鑒定出13個(gè)化合物,分別為蘋果酸(1)、枸櫞酸(2)、綠原酸(3)、花青素B2(4)、表兒茶素(5)、花青素B3(6)、牡荊素葡萄糖苷(7)、蘆?。?)、金絲桃苷(9)、異槲皮苷(10)、槲皮素(11)、熊果酸(12)、熊果醇(13)。結(jié)論 建立了一種簡(jiǎn)單、快速的UPLC/ESI-TOF/MS方法對(duì)山楂中化學(xué)成分進(jìn)行了鑒定。

[Abstract]
Objective To identify the chemical constituents of Crataegus pinnatifida var. major N. E. Brown by UPLC/ESI-TOF/MS. Methods The method of identification was performed on Acquity UPLC BEH C18 column (100 mm×2.1 mm, 1.7 μm), the mobile phase consisted of 0.1% formic acid aqueous solution and acetonitrile with gradient elution program. The injection volume was 1 μL at the flow rate of 0.3 mL/min. The column temperature was 50 ℃, and the detection wavelength was set from 260 to 360 nm. The detector was TOF Mass Spectrometer with an ESI ion source. The data were compared with those of the reference substances. Results Thirteen compounds were identified as malic acid (1), citric acid (2), chlorogenic acid (3), anthocyanin B2 (4), epicatechin (5), anthocyanin B3 (6), vitexin-glucoside (7), rutin (8), hyperoside (9), isoquercetin (10), quercetin (11), ursolic acid (12), and uvaol (13). Conclusion The established method for the determination of chemical constituents of C. pinnatifida var. major using UPLC/ESI- TOF/MS is simple and reliable.
