[摘要]
目的 研究血三七Polygonum amplexicaule D. Don var. sinense Forb.et Hemsl.ex Stew.水煎液對大鼠腦缺血再灌注損傷的保護作用,并探討其機制。方法 180只雄性SD大鼠隨機分為腦缺血再灌注(模型)組,假手術(shù)組,銀杏葉提取物(0.2 g/kg)組,血三七水煎液高、中、低劑量(0.3、0.2、0.1 g/kg)組,給藥組術(shù)前連續(xù)ig 14 d,于末次給藥60 min后行手術(shù),采用結(jié)扎頸外動脈建立大鼠大腦中動脈局灶性腦缺血再灌注損傷模型,觀察血三七水煎液組對腦缺血再灌注損傷大鼠神經(jīng)功能、腦梗死組織質(zhì)量比、腦含水量、腦組織中生化指標(biāo)的影響。結(jié)果 與模型組比較,血三七水煎液高、中劑量組,銀杏葉提取物組均可明顯改善神經(jīng)功能障礙,明顯降低腦梗死組織質(zhì)量比、減低腦組織中含水量,升高大鼠腦組織中SOD活性,降低MDA、NO的量(P<0.05、0.01)。結(jié)論 血三七水煎液對大鼠腦缺血再灌注損傷具有保護作用,其作用機制可能與其抗自由基和抑制NO生成有關(guān)。

[Abstract]
Objective To observe the protective effect of Polygonum amplexicaule var. sinense water decoction (PAWD) on focal cerebral ischemia-reperfusion (I/R) injury in rats, and to explore its mechanisms. Methods SD male rats (120 rats) were randomly divided into I/R, Sham, Ginkgo biloba extract (GBE, 0.2 g/kg), high-, mid-, and low-dose (0.3, 0.2, and 0.1 g/kg) PAWD groups. The animals were ig administered with drugs for 14 d. The operation was performed to induce the middle cerebral artery occlusion model in rats with the method of intraluminal thread at 60 min after the last administration via ligation of external carotid artery. After ischemia for 2 h and repefusion for 24 h in each model rat, the influences of PAWD on nerve function, tissue weight rate of cerebral infarction, water contents in brain, and biochemical indicators in brain were determined. Results Compared with I/R group, high-, mid-dose PAWD, and GBE could obviously improve the neurological dysfunction of rats, reduce the brain water contents, decrease tissue weight rate of cerebral infarction, increase the activities of SOD, and. reduce MDA and NO contents (P < 0.05 or 0.01). Conclusion PAWD has protective effect on cerebral I/R injury, and the mechanism may be related to attenuating free radical and NO.
