[摘要]
目的 研究大孔樹脂純化兩色金雞菊總黃酮的工藝條件及參數(shù)。方法 采用靜態(tài)吸附解吸法優(yōu)選適宜的大孔樹脂,以樹脂吸附量、總黃酮解吸率為考察指標,對篩選出的樹脂進行工藝優(yōu)化。結(jié)果 D-101樹脂具有較好的吸附及解吸附性能,其最佳工藝為:上樣液質(zhì)量濃度3.4 mg/mL,樹脂的上樣量為15 mL/g,上樣液pH值為5.0,上樣流速1.0 mL/min,6 BV去離子水洗脫除雜后,用pH 5.0的70%乙醇以1.5 mL/min洗脫5 BV;所得兩色金雞菊總黃酮質(zhì)量分數(shù)為73.5%,得率為8.8%。結(jié)論 D-101樹脂綜合性能好,適合于兩色金雞菊總黃酮的分離純化。

[Abstract]
Objective To explore the technological parameters and the purification process of total flavonoids from Coreopsis tinctoria. Methods Nine types of macroporous resins were compared through static adsorption-desorption methods with adsorbability and desorption ratio as indexes to optimize the purification parameters. Results D-101 resin showed better adsorption and desorption parameters, and the optimal parameters for purification process were as follows: the concentration of sample extract was 3.4 mg/mL, the loaded amount was 15 mL/g, pH value was 5.0, the flow rate was 1.0 mL/min, and after being eluted with 6 BV water, the optimal solution for desorption was 70% ethanol in pH 5.0 with 5 BV at the flow rate of 1.5 mL/min. The purity of total flavonoids was 73.5%, and the yield was 8.8%. Conclusion D-101 resin shows significant effect on the purification of flavonoids which is feasible to separate and purify total flavonoids from C. tinctoria.
