[摘要]
目的 比較銀杏內(nèi)生真菌在浙江天目山地區(qū)和建德地區(qū)以及在同一地區(qū)雄株與雌株銀杏中的種群結(jié)構(gòu)。方法 利用PDA培養(yǎng)基對采集自天目山地區(qū)和建德地區(qū)以及天目山地區(qū)雄株和雌株的銀杏內(nèi)生真菌分別進行分離、純化,根據(jù)菌株群落的培養(yǎng)特征及5.8S和ITS區(qū)片段序列相似性比較和系統(tǒng)發(fā)育分析,進行菌株鑒定。結(jié)果 從天目山地區(qū)和建德地區(qū)分別分離得到440、382株內(nèi)生真菌。天目山地區(qū)內(nèi)生真菌歸屬到9目、14科、19屬、28種;建德地區(qū)內(nèi)生真菌歸屬到8目、10科、11屬、26種。銀杏各組織部位內(nèi)生真菌的數(shù)量和種類按由多到少排序:枝條>樹皮>葉。天目山地區(qū)雄株銀杏內(nèi)生真菌在定殖率和菌株個數(shù)上均高于雌株銀杏。結(jié)論 銀杏內(nèi)生真菌具有極其豐富的物種多樣性,天目山地區(qū)銀杏內(nèi)生真菌在種類和數(shù)量上均多于建德地區(qū)。天目山地區(qū)雌雄株銀杏在菌種種類和數(shù)量上存在差異,雄株銀杏內(nèi)生真菌在菌種數(shù)量上具有一定的優(yōu)勢。

[Abstract]
Objective To compare endophytic fungi isolated from Ginkgo biloba in Tianmu Mountain and Jiande area of Zhejiang province, and to compare the male and female plant population structures in Tianmu Mountain. Methods PDA medium was used to separate and purify the endophytic fungi in G. biloba from Tianmu Mountain area and Jiande area, as well as male and female samples in Tianmu Mountain area. The collected endophytic fungi were identified by morphology, 5.8S, and ITS sequences. Results Endophytic fungi (440 strains) were isolated from Tianmu Mountain area and belonged to 28 species, 19 genera, 14 families, and 9 orders, and 382 Strains of endophytic fungi were isolated from Jiande area and belonged to 26 species, 11 genera, 10 families, and 8 orders. The amount and species of endophytic fungi in different tissues of G. biloba ranked from more to less as branch > bark > leaf. In G. biloba of Tianmu Mountain area, the endophytic fungi colonization rate and strain amount of male plants were higher than those in female plants. Conclusion Endophytic fungi from G. biloba show a great diversity. The endophytic fungi amount and species from G. biloba in Tianmu Mountain area are more than those in Jiande area. There are also differences in endophytic fungi amount and species between male and female G. biloba in Tianmu Mountain area.
