[摘要]
目的 探討紅景天與玄參組合物對局灶性腦缺血再灌注大鼠的保護作用。方法 通過體外活性評價,篩選出紅景天與玄參組合物最佳配比;采用頸內(nèi)動脈線栓法建立局灶性腦缺血模型,觀察模型大鼠的行為、腦梗死面積和腦含水量的變化;測定血清超氧化物歧化酶(SOD)活性和丙二醛(MDA)的含量。結(jié)果 當(dāng)紅景天與玄參以1∶1配比時,綜合評分最好;且組合物中劑量組(0.39 g/kg)、高劑量組(1.2 g/kg)均能顯著縮小局灶性腦缺血再灌注大鼠的腦梗死面積、降低梗死率,并能減少腦含水量(P<0.05),同時使腦缺血72 h大鼠的行為學(xué)癥狀得到顯著改善(P<0.05),明顯提高SOD活性并降低MDA的含量(P<0.05)。結(jié)論 紅景天與玄參組合物對局灶性腦缺血再灌注大鼠具有一定保護作用,且作用明顯優(yōu)于單用紅景天或玄參。

[Abstract]
Objective To investigate the protective effect of the combination of Rhodiolae Crenulatae Radix et Rhizomaand Scrophulariae Radix on focal cerebral ischemia-reperfusion injury of rats. Methods The in vitro activity evaluation was used to select the best ratio group of combination, and the internal carotid artery suture method was adopted to establish the model of focal cerebral ischemia model, and to observe the influence on the behavioral score, cerebral infarction area, and cerebral water content of model rats. Activity of superoxide dismutase (SOD) and content of malondialdehyde (MDA) in serum of rats were measured. Results The evaluation on the activity of the combination with the ratio in 1:1 was the best, and the combination in the middle dose (0.39 g/kg), high dose (1.2 g/kg) groups could reduce the infarction area in the cerebral ischemia-reperfusion rats, decrease the infarction rate, reduce brain water content significantly (P<0.05), at the same time, 72 h after modeling, The behavioral symptoms of the rats were improved significantly (P<0.05). The combination could increase the activity of SOD and reduce the content of MDA (P<0.05). Conclusion The combination of Rhodiolae Crenulatae Radix et Rhizomaand Scrophulariae Radix has a certain protective effect on the focal cerebral ischemia-reperfusion injury of rats, and the effect of the combination is superior to Rhodiolae Crenulatae Radix et Rhizoma alone or Scrophulariae Radix.
