[摘要]
目的 建立去甲斑蝥素體外肝細(xì)胞代謝產(chǎn)物的HPLC-MS分析方法。方法 去甲斑蝥素進(jìn)行體外肝細(xì)胞代謝,透析得到代謝產(chǎn)物,并用HPLC-MS對(duì)產(chǎn)物進(jìn)行鑒定,再通過半制備HPLC色譜儀分離純化,最后根據(jù)理化性質(zhì)和波譜數(shù)據(jù)鑒定代謝物的結(jié)構(gòu)。結(jié)果 共分離得到7個(gè)化合物,對(duì)其中量較高的2個(gè)化合物進(jìn)行結(jié)構(gòu)鑒定,分別為去甲斑蝥素與谷氨酸結(jié)合物(1)、去甲斑蝥酸鈉(2)。結(jié)論 體外細(xì)胞代謝法由于反應(yīng)條件相對(duì)單純,易于控制,適合于制備去甲斑蝥素體外肝細(xì)胞代謝產(chǎn)物。

[Abstract]
Objective To identify the liver cell metabolites of norcantharidin in vitro by HPLC-MS. Methods The norcantharidin metabolized by liver cell in vitro, then the metabolites were obtained by dialysis, and the metabolites were identified. Then they were separated and purified by semi-preparing HPLC. Their structures were identified on the basis of physicochemical properties and spectroscopic data.Results Seven compounds were separated and two compounds with higher contents were identified as combination of norcantharidin and glutamic acid (1), and sodium demethylcantharidate (2). Conclusion The reaction conditions of cell metabolism in vitro are relatively simple and easy to control, therefore the method is suitable for the preparation of liver cell metabolites of norcantharidin in vitro.
