[摘要]
目的 探討側(cè)腦室注射apelin-13對小鼠體溫的作用及對脂多糖誘導(dǎo)小鼠發(fā)熱的解熱作用。方法 小鼠隨機(jī)分為對照組,脂多糖50、150 ng組,apelin-13 3、10 nmol組,脂多糖150 ng+apelin-13 3 nmol組和脂多糖150 ng+apelin-13 10 nmol組。藥物或生理鹽水以10 μL/min的恒定速度側(cè)腦室注射,注射體積為4 μL。給藥后0.5、1.0、1.5、2.0、2.5、3.0、3.5、4.0 h分別監(jiān)測記錄各組小鼠的體溫,計算各組小鼠在各時間點體溫的變化。根據(jù)實驗結(jié)果繪制溫度變化曲線,并計算出曲線下面積(area under the curve,AUC)。結(jié)果 與對照組比較,脂多糖150 ng組溫度均顯著升高(P<0.05),引起發(fā)熱效果明顯,因此選擇注射150 ng脂多糖作為模型組。側(cè)腦室單獨注射3、10 nmol apelin-13對小鼠溫度變化沒有明顯影響。側(cè)腦室注射3 nmol apelin-13在1.5 h降低150 ng脂多糖引起的發(fā)熱(P<0.05);而10 nmol apelin-13在1.5、2.0、3.0 h均降低150 ng脂多糖引起的發(fā)熱(P<0.05)。與脂多糖150 ng組比較,脂多糖150 ng+apelin-13 10 nmol組體溫AUC顯著減少(P<0.05);脂多糖150 ng+apelin-13 3 nmol組體溫AUC雖然減少,但是并不具有顯著性差異。結(jié)論 中樞apelin-13對小鼠的體溫沒有影響,但側(cè)腦室注射10 nmol apelin-13能夠明顯降低脂多糖引起的小鼠體溫升高,具有緩解脂多糖誘導(dǎo)的小鼠發(fā)熱作用。

[Abstract]
Objective To study the influence of introcerebroventricular (icv) administered with apelin-13 on body temperature of mice, and investigate antipyretic effect on lipopolysaccharide (LPS)-induced fever in mice. Methods Mice were randomly divided into control group, LPS 50 and 150 ng groups, apelin-13 3 and 10 nmol groups, LPS 150 ng + apelin-13 3 nmol group, and LPS 150 ng + apelin-13 10 nmol group. Drugs or normal saline were icv administered at a constant speed of 10 L/min, and the injection volume was 4 μL. Body temperature was monitored and recorded at the point of 0.5, 1.0, 1.5, 2.0, 2.5, 3.0, 3.5, and 4.0 h after treatment. The changes of body temperature in mice were calculated. Temperature change curve was drawn according to the experiment, and area under the curve (AUC) was also calculated. Results Compared with the control group, body temperature in LPS 150 ng group was significantly increased (P < 0.05), so it was selected as model group in the next experiment. There were no changes of body temperature in mice after icv administered with apelin-13 3 and 10 nmol. Apelin-13 could decrease 150 ng LPS-induced fever in mice with the dosage of 3 nmol during 1.5 h (P < 0.05), and also with the dosage of 10 nmol during 1.5, 2.0, and 3.0 h (P < 0.05). Compared with the LPS 150 ng group, the change of AUC of body temperature in LPS 150 ng + apelin-13 10 nmol group was significantly decreased (P < 0.05), but that in LPS 150 ng + apelin-13 3 nmol group had no significant differences. Conclusion Apelin-13 in central system has no effect on the body temperature of mice. But icv 10 nmol apelin-13 can relieve the body temperature of mice with LPS-induced fever, and has antipyretic effect on LPS-induced fever in mice.
