2腎上腺素能受體(β2-AR)探討其作用機(jī)制。方法 大鼠隨機(jī)分為對(duì)照組、模型組、沙丁胺醇組、普萘洛爾組、白及組、附子組和反藥組合組。乙酸貼敷法制備胃潰瘍模型,ig給藥,觀察潰瘍面積、胃黏膜病理形態(tài)變化。利用Elisa試劑盒檢測(cè)胃組織勻漿炎癥相關(guān)指標(biāo)髓過氧化物酶(MPO)、白細(xì)胞介素6(IL-6)、環(huán)磷酸腺苷(cAMP)水平變化。采用雙熒光素酶報(bào)告基因技術(shù)評(píng)價(jià)附子、反藥組合對(duì)β2-AR的激動(dòng)作用。結(jié)果 與模型組比較,附子組、反藥組合組潰瘍嚴(yán)重、自愈不佳,差異有統(tǒng)計(jì)學(xué)意義(P<0.05);白及組潰瘍較輕,愈合明顯,差異有極顯著意義(P<0.01)。與模型組比較,反藥組合組IL-6、MPO、cAMP水平均顯著升高(P<0.05、0.01),附子組MPO水平顯著升高(P<0.01)。附子具有明顯的激動(dòng)β2-AR作用,與白及聯(lián)合使用后激動(dòng)作用進(jìn)一步增強(qiáng)(P<0.01)。結(jié)論 附子、白及反藥組合加重胃潰瘍和炎癥反應(yīng)是其“相反”的主要表現(xiàn),其機(jī)制可能與協(xié)同激動(dòng)β2腎上腺素能受體有關(guān)。;Objective To observe toxic effect performance of Aconiti Lateralis Radix Preparata and Bletillae Rizoma anti drug combination under the conditions of acetic acid induced gastric ulcer in rats, and explore its mechanism based on β2 adrenergic receptor (β2-AR). Methods Rats were randomly divided into normal group, model group, salbutamol group, propranolol group, Aconiti Lateralis Radix Preparata group, Bletillae Rizoma group, and anti drug combination group. Acetic acid was applied to make gastric ulcer model, and area of gastric ulcer and pathological morphological changes were observed after filling the stomach with medicines. Indicators related with homogenate inflammation MPO, IL-6, and cAMP changes in stomach tissue were determined by Elisa kits method. Activating actions on β2-AR of Aconiti Lateralis Radix Preparata and Bletillae Rizoma anti drug combination were evaluated with technology of double luciferase report gene. Results Compared with model group, ulcer of Aconiti Lateralis Radix Preparata and anti drug combination group were serious, the self healings were not good (P<0.05), but ulcers of Bletillae Rizoma group were light, and the healings were obvious (P<0.01). Compared with model group, levels of IL-6, MPO, and cAMP in anti drug combination group were significantly increased (P<0.05, 0.01), and levels of MPO in Aconiti Lateralis Radix Preparata group were also significantly increased (P<0.01). Aconiti Lateralis Radix Preparata had obvious activating action on β2-AR, and this action had further enhanced after combination with Bletillae Rizoma (P<0.01).
