[摘要]
目的 評價五味子不同炮制品對小鼠慢性不可預(yù)知性應(yīng)激抑郁的改善作用。方法 ICR小鼠隨機分為對照組、模型組、氟西汀組以及生五味子組、酒五味子組、醋五味子組,每組10只。對照組和模型組ig蒸餾水,氟西汀組ig鹽酸氟西汀分散片3.3 mg/kg,生五味子、酒五味子和醋五味子組均ig相應(yīng)藥物1.0 g/kg,體積為10 mL/kg,給藥1 h后采用10種刺激方法建立慢性不可預(yù)知性應(yīng)激模型,持續(xù)28 d。以敞箱實驗、懸尾實驗、強迫游泳實驗進(jìn)行行為學(xué)觀察,采用酶聯(lián)免疫法測定血清中五羥色胺(5-HT)、多巴胺(DA)、腎上腺皮質(zhì)酮(CORT)水平,對實驗結(jié)果進(jìn)行綜合分析。結(jié)果 與模型組比較,生五味子可以增加抑郁小鼠敞箱站立次數(shù)(P<0.01)、敞箱中央格穿越次數(shù)(P<0.05),降低懸尾、強迫游泳不動時間(P<0.05);酒五味子和醋五味子均對抑郁小鼠敞箱站立次數(shù)、敞箱中央格穿越次數(shù)無影響,但可降低懸尾不動時間、強迫游泳不動時間(P<0.05)。與模型組比較,生五味子可顯著升高抑郁小鼠血清中5-HT、DA水平,降低CORT水平(P<0.01);酒五味子和醋五味子可顯著升高抑郁小鼠血清中DA水平,降低CORT水平(P<0.05、0.01),對血清中5-HT水平無影響。結(jié)論 五味子不同炮制品均有一定的抗抑郁作用,其中生五味子作用較強。

[Abstract]
Objective To evaluate the improvement of Schisandrae Fructus processed products on depression of mice induced by chronic unpredictable mild stress. Methods ICR mice were randomly divided into control group, model group, fluoxetine group, and Schisandrae Fructus, Schisandrae Fructus processed with alcohol, and Schisandrae Fructus processed with wine groups, and each group had 10 mice. Mice in the control and model groups were ig administered with distilled water. Mice in the fluoxetine group were ig administered with Fluoxetine Hydrochloride Dispersible Tablets 3.3 mg/kg. Mice in the Schisandrae Fructus groups were ig administered with Schisandrae Fructus 1.0 g/kg with volume of 10 mL/kg. Ten kinds of unpredictable stimulations were adopted to establish the model of chronic unpredictable mild stress after last administration 1 h for 28 d. Behaviors of depressive mice in open field test, tail suspression test, and forced swimming test were observed. The contents of 5-HT, DA, and CORT in serum were determined by Enzyme-linked immunosorbent assay (ELISA) method. Results were analyzed by comprehensive analysis method. Results Compared with the model group, the open field stand times and open central grid crossing times were increased, but the time of hanging tail and room temperature forced swimming were decreased in the Schisandrae Fructus group (P<0.05, 0.01). There were no differences on open field stand times and open central grid crossing times in Schisandrae Fructus processed with alcohol and wine groups, but the time of hanging tail and room temperature forced swimming were decreased (P<0.05). Compared with the model group, contents of 5-HT and DA in serum of depressive mice were increased, but the contents of CORT in serum were decreased in the Schisandrae Fructus group (P<0.01). The contents of 5-HT in serum of depressive mice were increased, but the contents of CORT in serum were decreased in the Schisandrae Fructus processed with alcohol and wine groups (P<0.05, 0.01), but there had no differences on contents of DA in serum. Conclusion Schisandrae Fructus processed products have antidepressant effect on depression of mice induced by chronic unpredictable mild stress, and Schisandrae Fructus has stronger effect in them.
