18柱色譜柱(250 mm×4.6 mm,5 μm);流動(dòng)相:乙腈–0.4%磷酸溶液,梯度洗脫;0~14 min時(shí)在280 nm波長(zhǎng)下檢測(cè)熊果苷,14~18 min在350 nm波長(zhǎng)下檢測(cè)槲皮苷,18~30 min在256 nm波長(zhǎng)下檢測(cè)纈草三酯和乙酰纈草三酯;體積流量:0.9 mL/min;柱溫:25 ℃;進(jìn)樣量為10 μL。結(jié)果 熊果苷、槲皮苷、纈草三酯和乙酰纈草三酯分別在11.97~239.40 μg/mL、4.65~93.00 μg/mL、4.18~83.60 μg/mL、5.30~106.00 μg/mL峰面積與質(zhì)量濃度呈良好的線性關(guān)系,r分別為0.999 8、0.999 3、0.999 9、0.999 5;平均回收率分別為98.78%、97.11%、99.14%、98.75%,RSD值分別為1.62%、0.68%、1.39%、0.87%。結(jié)論 所建立的方法樣品處理方法簡(jiǎn)便,檢測(cè)的準(zhǔn)確性和重復(fù)性均較好,可為醒脾養(yǎng)兒顆粒質(zhì)量標(biāo)準(zhǔn)測(cè)定方法的制定提供依據(jù)。;Objective To develop an HPLC wavelength switching method for simultaneous determination of the contents of arbutin, quercitrin, valepotriate and acevaltrate in Xingpi Yang'er Granules. Methods HPLC method was adopted. The determination was carried out on Agilent HC-C18 chromatographic column (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitril - 0.4% phosphoric acid solution with gradient elution. The detection wavelengths were set at 280 nm in 0 — 14 min (determination of arbutin), 350 nm in 14 — 18 min (determination of quercitrin), and 256 nm in 18 — 30 min (determination of valepotriate and acevaltrate). The flow rate was 0.9 mL/min, temperature of column was set at 25 ℃, and volume of injection was 10 μL. Results The linear range ofarbutin, quercitrin, valepotriate and acevaltrate were 11.97 — 239.40 μg/mL (r=0.999 8), 4.650 — 93.00 μg/mL (r=0.999 3), 4.180 — 83.60 μg/mL (r=0.999 9), and 5.300 — 106.0 μg/mL (r=0.999 5), respectively. The average recoveries were 98.78%, 97.11%, 99.14%, and 98.75% with RSD 1.62%, 0.68%, 1.39%, and 0.87%, respectively. Conclusion The established method is simple and accurate, has good repeatability which can be applied to the quality control for Xingpi Yang'er Granules."/>
