[摘要]
目的 探索芫花素對于MC3T3-E1細胞向成骨細胞分化的影響。方法 采用芫花素1、5、10、15 μmol/L處理MC3T3-E1細胞14 d,采用MTS法檢測芫花素對MC3T3-E1細胞增殖的抑制作用;實時定量PCR和Western blotting法檢測測芫花素對MC3T3-E1細胞中ALP、HDAC1和Runx2 mRNA和蛋白表達的影響。結(jié)果 芫花素5、10、15 μmol/L可以顯著促進MC3T3-E1細胞中ALP、Runx2 mRNA和蛋白表達水平的提高(P<0.05),HDAC1 mRNA和蛋白表達水平的下調(diào)(P<0.05),表明芫花素可以促進MC3T3-E1向成骨細胞分化。結(jié)論 芫花素具有促進MC3T3-E1細胞可以促進成骨細胞分化,對成骨細胞的分化過程有一定的調(diào)節(jié)作用。

[Abstract]
Objective To study the effect of genkwanin on the differentiation of MC3T3-E1 cell into osteoblast. Methods The MC3T3-E1 cell was treated with genkwanin 1, 5, 10, and 15 μmol/L for 14 d. Inhibition of genkwanin on proliferation of MC3T3-E1 cell was detected by MTS method. Effect of genkwanin on ALP、HDAC1, and Runx2 mRNA and protein expressions of MC3T3-E1 cell was detected by the real-time quantitative PCR and Western blotting. Results Genkwanin 5, 10, and 15 μmol/L could promote the expression of ALP and Runx2 mRNA and protein expressions in MC3T3-E1 cells (P<0.05), but inhibit the expression of HDAC1 mRNA and protein expressions in MC3T3-E1 cells (P<0.05). Conclusion Genkwanin has the action of promoting the differentiation of MC3T3-E1 cell into osteoblasts, and can regulate the differentiation process of osteoblasts.
