目的 觀察蜂毒注射液對(duì)人類(lèi)風(fēng)濕關(guān)節(jié)炎成纖維樣滑膜細(xì)胞(類(lèi)風(fēng)濕關(guān)節(jié)炎-FLS)B淋巴細(xì)胞瘤-2基因(Bcl-2)、Bcl-2相關(guān)X蛋白(Bax)mRNA和蛋白的影響,探討蜂毒注射液治療類(lèi)風(fēng)濕關(guān)節(jié)炎的作用機(jī)制。方法 滑膜組織取自行關(guān)節(jié)置換或關(guān)節(jié)鏡手術(shù)的4名活動(dòng)期類(lèi)風(fēng)濕關(guān)節(jié)炎患者,分離出FLS并原代培養(yǎng),逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)觀察Bax、Bcl-2 mRNA表達(dá)的變化,酶聯(lián)免疫吸附測(cè)定(ELISA)法檢測(cè)Bax、Bcl-2蛋白水平的變化。結(jié)果 與對(duì)照組比較,蜂毒注射液4、8、16 mg/L組能促進(jìn)Bax mRNA表達(dá)(P<0.05),蜂毒注射液8、16 mg/L組抑制Bcl-2 mRNA表達(dá)(P<0.05、0.01);蜂毒注射液4、8、16 mg/L組提高Bax蛋白水平,降低Bcl-2蛋白水平(P<0.05、0.01),且有一定的劑量相關(guān)性。結(jié)論 蜂毒注射液可能通過(guò)抑制Bcl-2蛋白、促進(jìn)Bax蛋白的表達(dá),達(dá)到促進(jìn)類(lèi)風(fēng)濕關(guān)節(jié)炎-FLS凋亡的作用。

[Abstract]
Objective To observe the effect of Apistoxin Injection on B-lymphoma-2 gene (Bcl-2), Bcl-2-related X protein (Bax) mRNA and protein in human rheumatoid arthritis fibroblast like synoviocytes (RA-FLS), and to explore the mechanism of Apistoxin Injection in treatment of rheumatoid arthritis. Methods FLS was isolated from synovium of 4 active RA patients who underwent arthroscopic surgery or joint replacement. The expression of Bax and Bcl-2 mRNA was observed by Real time-PCR (RT-PCR) method, and the contents of Bax and Bcl-2 protein were detected by ELISA method. Results Compared with the control group, Apistoxin Injection 4, 8, and 16 mg/L groups could promote the expression of Bax mRNA (P < 0.05), and Apistoxin Injection 8 and 16 mg/L groups could inhibit the expression of Bcl-2 mRNA (P < 0.05, 0.01). Apistoxin Injection 4, 8, and 16 mg/L groups could increase the content of Bax protein and decrease the content of Bcl-2 protein (P < 0.05, 0.01), which was dose-dependent. Conclusion Apistoxin injection may promote the apoptosis of RA-FLS by inhibiting the expression of Bcl-2 protein and promoting the expression of Bax protein.
