[摘要]
目的 建立一種用于大鼠靜脈血栓模型的靜脈栓塞分級(jí)評(píng)價(jià)體系,為溶栓藥物用于臨床的適應(yīng)癥提供參考。方法 雄性Wistar大鼠制備靜脈血栓。在血栓形成后30 min開始給藥,采用尿激酶局部輸注41、82、164 kU/kg及阿替普酶2.05、4.10、8.20 mg/kg靜脈血栓進(jìn)行治療,給藥后3 h采用血管造影法拍攝靜脈栓塞部位的X光片,對靜脈栓塞進(jìn)行分級(jí)研究。結(jié)果 尿激酶、阿替普酶均產(chǎn)生劑量相關(guān)性溶栓作用,5級(jí)研究體系的研究結(jié)果與血管再通率,血管通過率、血栓質(zhì)量的結(jié)果一致。5級(jí)評(píng)分標(biāo)準(zhǔn),級(jí)距設(shè)計(jì)合理,在血栓殘留量、血管通過率方面顯示出明顯的級(jí)間差別。結(jié)論 基于靜脈造影的靜脈栓塞分級(jí)評(píng)價(jià)體系可客觀表現(xiàn)靜脈栓塞程度,可用于評(píng)估溶栓藥物靜脈溶栓效果。

[Abstract]
Objective To establish a system for grading evaluation system for venous thrombosis in rats, and to provide a reference for clinical indications of thrombolytic drugs. Methods Venous thrombosis models were made in male Wistar rats. Urokinase local infusion of 41, 82, 164 kU/kg, and ateplase of 2.05, 4.10, and 8.20 mg/kg at 30 min after thrombosis were used to treat venous thrombosis. After be treated for 3 h, the X-ray film of the site of venous embolism was taken by angiography to study the grade of venous embolism. Results Urokinase and ateplase produced a dose-dependent thrombolytic effect. Grade 5 system was consistent with those of recanalization rate, vascular throughput rate, and thrombus weight. The design of grade 5 was reasonable, and there were significant differences in thrombus residue and vascular throughput rate between each grade. Conclusion The grading evaluation system of venous embolism based on venography can objectively show the degree of venous embolism, and can be used to evaluate the thrombolytic effect of thrombolytic drugs.
