炎癥反應是機體常見的生理、病理活動,炎癥小體在該反應中發(fā)揮重要調控作用。核苷酸結合寡聚化結構域樣受體蛋白3(NOD-like receptor protein 3,NLRP3)是炎癥小體中關鍵的調控蛋白之一。研究發(fā)現激活NLRP3的刺激因子及其相關分子調控信號通路與多種疾病的發(fā)生、發(fā)展密切相關,并越來越受到廣泛關注,是臨床藥物研究開發(fā)的前沿熱點方向。總結NLRP3炎癥小體的生物結構、調控功能、信號通路和作用機制等研究進展,并梳理與其相關靶點抑制劑的研究結果,對開展炎癥相關疾病的新靶標藥物發(fā)現及其臨床應用研究具有重要的參考價值。

[Abstract]
The inflammatory response is a common physiological and pathological process of the body, which is generally induced by the activation of various inflammatory factors. NLRP3 is a key regulatory protein. It has been found that there are a variety of stimulating factors and molecular regulatory signal pathways involved in NLRP3 activation closely related to the occurrence and development of a variety of diseases, and have attracted more and more attention. It is one of the frontier hot topics in clinical drug research and development. This paper summarizes the research progress in biological structure, regulatory pathway and inhibitors development of NLRP3 inflammasome, which is helpful for the target seeking and clinical drug development.
