[摘要]
目的 運(yùn)用網(wǎng)絡(luò)藥理學(xué)方法探討祛痹痛風(fēng)飲治療痛風(fēng)的活性成分、作用靶點(diǎn)及其潛在的治療作用機(jī)制。方法 基于中藥系統(tǒng)藥理學(xué)數(shù)據(jù)庫與分析平臺(tái)(TCMSP)獲取祛痹痛風(fēng)飲的活性成分和作用靶點(diǎn),結(jié)合Uniprot數(shù)據(jù)庫對(duì)靶點(diǎn)進(jìn)行標(biāo)準(zhǔn)化處理。采用Cytoscape 3.7.2軟件繪制出祛痹痛風(fēng)飲的活性成分-痛風(fēng)靶點(diǎn)網(wǎng)絡(luò)。通過GeneCards、OMIM、TTD、DRUGBANK數(shù)據(jù)庫獲得痛風(fēng)的疾病靶點(diǎn)。將藥物的作用靶點(diǎn)與疾病靶點(diǎn)進(jìn)行交集,獲得共同靶點(diǎn)。將交集靶點(diǎn)應(yīng)用STRING數(shù)據(jù)庫構(gòu)建蛋白-蛋白相互作用網(wǎng)絡(luò)圖(PPI),通過DAVID數(shù)據(jù)庫對(duì)共同靶點(diǎn)進(jìn)行基因本體(GO)功能富集分析和京都基因與基因組百科全書(KEGG)通路富集分析,并將結(jié)果進(jìn)行可視化處理。結(jié)果 共預(yù)測(cè)到祛痹痛風(fēng)飲中191個(gè)活性成分,87個(gè)與痛風(fēng)共同的靶點(diǎn)。GO功能富集分析顯示與炎癥反應(yīng)、細(xì)胞對(duì)脂多糖的應(yīng)答、基因表達(dá)的正調(diào)控等相關(guān)。KEGG通路富集分析結(jié)果顯示主要涉及Toll樣受體信號(hào)通路、NOD樣受體信號(hào)通路、NF-κB信號(hào)通路等方面。結(jié)論 祛痹痛風(fēng)飲可能通過促進(jìn)尿酸排泄、抑制炎癥因子的釋放減輕、炎癥反應(yīng)發(fā)揮治療痛風(fēng)的作用,為深入研究祛痹痛風(fēng)飲治療痛風(fēng)的作用機(jī)制提供了理論參考。

[Abstract]
Objective To study the active ingredient, target and possible mechanism of Qubi Tongfeng Yin in treatment of gout by network pharmacological method. Methods TCMSP database was used to obtain the active ingredient and target of Qubi Tongfeng Yin, and the target was standardized by Uniprot database. Cytoscape3.7.2 software was used to draw the network of drug active ingredient and target of gout. The disease targets of gout were obtained through Genecards, OMIM, TTD, and DRUGBANK databases. The drug target is intersected with the disease target to obtain the drug and disease common target. PPI was constructed for the intersection targets using String database. Gene ontology (GO) biological process analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis were performed for the common targets using David database, and the results were visualized. Results A total of 191 active components of Qubi Tongfeng Yin were predicted in this study, and 87 common targets of Qubi Tongfeng Yin were predicted. The results of GO biological process showed that it was related to inflammatory response, cellular response to lipopolysaccharide, and positive regulation of gene expression. KEGG pathway enrichment analysis showed that it mainly involved Toll-like receptor signaling pathway, NOD-like receptor signaling pathway, NF-κB signaling pathway and so on. Conclusion Qubi Tongfeng Yin may play a role in treatment of gout by promoting uric acid excretion and inhibiting the release of inflammatory factors to reduce inflammatory response, which provides a theoretical basis for the further study of the mechanism of Qubi Tongfeng Yin in treatment of gout.
