[摘要]
目的 通過網(wǎng)絡藥理學方法篩選金銀花-連翹藥對中活性成分和IgA腎病靶點,探討金銀花-連翹藥對干預IgA腎病的作用機制。方法 采用中藥系統(tǒng)藥理學數(shù)據(jù)庫(TCMSP)檢索金銀花、連翹中活性成分及其作用靶點。通過OMIM、NCBI、GeneCards數(shù)據(jù)庫獲取IgA腎病相關基因靶點。利用Venny 2.1軟件得到中藥活性成分作用靶點與IgA腎病的交集靶點信息。利用String數(shù)據(jù)庫和Cytoscape 3.8.0制作共有靶點蛋白相互作用網(wǎng)絡圖。運用R語言對核心作用靶點進行GO富集分析和KEGG通路分析。結果 金銀花-連翹藥對作用于IgA腎病的主要活性成分有槲皮素、毛地黃黃酮、山柰酚、β-胡蘿卜素、β-谷甾醇;作用的疾病靶點有AKT1、IL6、MAPK1、JUN、VEGFA、MAPK8、IL1B、MMP9;主要通過對化學應激、氧化應激、脂多糖、細菌來源分子的反應等生物過程和AGE-RAG、IL-17、TNF信號通路發(fā)揮作用。結論 金銀花-連翹藥對干預IgA腎病通過多成分、多靶點、多通路發(fā)揮作用,通過網(wǎng)絡藥理學方法為二藥的機制研究提供了數(shù)據(jù)支持。

[Abstract]
Objective To study the active components and mechanism of Lonicerae Japonicae Flos and Forsythiae Fructus in the treatment of IgA nephropathy based on network pharmacology. Methods TCMSP database was used to extract the active components and related targets of Lonicerae Japonicae Flos and Forsythiae Fructus. The disease targets were searched by OMIM, NCBI and GeneCards database. The information of the intersection targets of the active components of Lonicerae Japonicae Flos, Forsythiae Fructus, and IgA nephropathy were obtained by using Venny2.1 software. PPI network of the intersection target of Lonicerae Japonicae Flos, Forsythiae Fructus, and IgA nephropathy was draw by String and Cytoscape 3.8.0. The R language was used to perform GO enrichment analysis and KEGG pathway analysis.Results The study showed that the main active components include quercetin, luteolin, kaempferol, beta-carotene, and beta-sitosterol. The disease targets include AKT1, IL6, MAPK1, JUN, VEGFA, MAPK8, IL1B, and MMP9. The main biological process include cellular response to chemical stress, response to oxidative stress, response to lipopolysaccharide, and response to molecule of bacterial origin. The treating signaling pathway include AGE-RAGE signaling pathway, IL-17 signaling pathway, and TNF signaling pathway. Conclusion Lonicerae Japonicae Flos and Forsythiae Fructus treating IgA nephropathy are through multi-component, multi-target, and multi-signaling pathway, which provides data support for the further study of the two herbs based on network pharmacology.
