[摘要]
目的 研究牛樟葉總多糖對(duì)大鼠慢性酒精性肝損傷的保護(hù)作用與機(jī)制。方法 60只雄性大鼠按體質(zhì)量隨機(jī)分為對(duì)照組、模型組、多烯磷脂酰膽堿膠囊(10.944 mg/kg)組及牛樟葉總多糖高、中、低劑量(50.0、37.5、25.0 mg/kg)組。對(duì)照組和模型組給予蒸餾水,其余各組給予不同劑量藥物。給藥0.5 h后,除對(duì)照組外,其余各組均ig體積分?jǐn)?shù)為56%紅星二鍋頭,共給藥8周。采用蘇木精-伊紅(HE)染色觀察各組肝組織形態(tài)及結(jié)構(gòu)的變化,測(cè)定各組大鼠血清白細(xì)胞介素-1β(IL-1β)、IL-6、腫瘤壞死因子-α(TNF-α)、丙氨酸基轉(zhuǎn)移酶(ALT)、谷氨酸基轉(zhuǎn)移酶(AST)、三酰甘油(TG)及肝組織勻漿液中IL-1β、IL-6、TNF-α、谷胱甘肽(GSH)水平。免疫組織化學(xué)法測(cè)定大鼠肝臟沉默信息調(diào)節(jié)因子1(SIRT1)、磷酸化腺苷酸活化蛋白激酶α(p-AMPKα)表達(dá)水平。蛋白質(zhì)印跡法測(cè)定肝組織SIRT1、AMPKα、p-AMPKα、甾醇調(diào)節(jié)元件結(jié)合蛋白1c(SREBP1c)水平。結(jié)果 病理切片結(jié)果表明,牛樟葉總多糖能改善大鼠肝組織病理變化。與模型組相比,牛樟葉總多糖各劑量組IL-1β、IL-6、TNF-α、TG、ALT、AST水平均明顯降低(P<0.05);GSH水平明顯升高(P<0.05)。牛樟葉總多糖可提高SIRT1表達(dá)及AMPKα的磷酸化水平。結(jié)論 牛樟葉總多糖對(duì)慢性酒精性肝損傷有一定的保護(hù)作用,其機(jī)制可能與抗氧化應(yīng)激、抑制炎癥反應(yīng)、調(diào)節(jié)SIRT1/AMPK信號(hào)通路有關(guān)。

[Abstract]
Objective To study the protective effect and mechanism of polysaccharide from Cinnamomum camphora leaves on chronic alcoholic liver injury in rats. Methods Sixty male rats were randomly divided into control group, model group, Polyene Phosphatidylcholine Capsules (10.944 mg/kg), polysaccharide from C. camphora leaves high-, medium- and high-dose (50, 37.5 and 25mg/kg) groups according to body weight. Rats in the control group and model group was given distilled water, and rats in the other groups was given corresponding drugs regularly every day. In addition, rats in the treatment groups and model group were given 56% red star erguotou by intragastric administration 0.5 h later to establish chronic alcoholic liver injury model. They were treated for 8 weeks. Hematoxylin-eosin (HE) staining was used to observe the changes of liver tissue morphology and structure in each administration group. The levels of IL-1β, IL-6, TNF-α, ALT, AST, and TG in serum and IL-1β, IL-6, TNF-α, and GSH in liver tissue homogenate were determined. The protein levels of SIRT1, AMPKα, P-AMPK α and SREBP1c in liver tissue were determined by Western blotting. The expression levels of SIRT1 and P-AMPK α in rat liver were determined by immunohistochemistry. Results Pathological section results showed that polysaccharide from C. camphora leaves could improve the pathological changes of rat liver tissue. Compared with model group, the levels of IL-1β, IL-6, TNF-α, TG, ALT and AST in all dose groups were significantly decreased (P < 0.05), while the level of GSH was significantly increased (P < 0.05). Total polysaccharide from C. camphora leaves can increase the expression of SIRT1 and phosphorylation of AMPKα. Conclusion Total polysaccharide from C. camphora leaves has a certain protective effect on chronic alcoholic liver injury, and its mechanism may be related to antioxidant stress, anti-inflammatory response and regulation of SIRT1-AMPK signaling pathway.
