[摘要]
目的 探究藍(lán)萼乙素通過蛋白激酶B(Akt)/B細(xì)胞淋巴瘤-2(Bcl-2)相關(guān)死亡啟動(dòng)因子(BAD)通路對(duì)宮頸癌裸鼠移植瘤生長(zhǎng)的影響和機(jī)制。方法 將48只SPF級(jí)雌性Balb/c-nu裸鼠隨機(jī)分為模型、藍(lán)萼乙素(56、112 mg/kg)和藍(lán)萼乙素+SC79(112 mg/kg+10 mg/kg)組,每組12只。將HeLa細(xì)胞種植在裸鼠右大腿根部背側(cè)皮下,建立皮下移植瘤模型,觀察腫瘤體積、質(zhì)量和一般情況的變化。蘇木精–伊紅(HE)染色觀察腫瘤組織的組織學(xué)變化。TUNEL實(shí)驗(yàn)檢測(cè)腫瘤組織細(xì)胞凋亡情況。Western blotting法檢測(cè)移植瘤組織中p-Akt、Akt、p-BAD、BAD、磷酸酯酶與張力蛋白同源物(PTEN)蛋白表達(dá)水平。結(jié)果 與模型組相比,藍(lán)萼乙素組第22、25、28天裸鼠腫瘤體積、腫瘤質(zhì)量、腫瘤組織Akt、BAD磷酸化水平顯著降低(P<0.05),腫瘤細(xì)胞凋亡率、腫瘤組織PTEN蛋白水平顯著升高(P<0.05);SC79可減弱藍(lán)萼乙素對(duì)宮頸癌裸鼠移植瘤生長(zhǎng)的抑制作用。結(jié)論 藍(lán)萼乙素對(duì)宮頸癌裸鼠移植瘤生長(zhǎng)起到抑制作用,其作用機(jī)制與抑制Akt/BAD信號(hào)通路有關(guān)。

[Abstract]
Objective To investigate the impact and mechanism of glaucocalyxin B on the growth of cervical cancer transplanted tumor in nude mice through the Akt/Bcl-2 associated BAD pathway. Methods 48 SPF grade female Balb/c nu nude mice were randomly grouped into model group, glaucocalyxin B (56, 112 mg/kg), and glaucocalyxin B + SC79 (112 mg/kg + 10 mg/kg) group, with 12 mice in each group. HeLa cells were implanted subcutaneously at the dorsal side of the right thigh root in nude mice to establish a subcutaneous transplanted tumor model, and the changes in tumor volume, weight, and general situation were observed. HE staining was applied to observe the pathological changes of tumor tissue. TUNEL assay was applied to detect the apoptosis of tumor histiocyte. Western blotting method was applied to detect the expression levels of p-Akt, Akt, p-BAD, BAD, and PTEN proteins in transplanted tumor tissue. Results Compared with the model group, the tumor volume at 22, 25, and 28 day, tumor weight, Akt, and BAD phosphorylation levels in tumor tissue of the glaucocalyxin B group were obviously decreased (P < 0.05), the apoptosis rate of tumor cells and the expression of PTEN protein in tumor tissue were obviously increased (P < 0.05). SC79 could weaken the inhibitory effect of glaucocalyxin B on the growth of cervical cancer transplanted tumor in nude mice. Conclusion Glaucocalyxin B inhibited the growth of transplanted tumors in nude mice with cervical cancer, and its mechanism of action was related to the inhibition of the Akt/BAD signaling pathway.
