[摘要]
目的 采用超高效液相色譜-質(zhì)譜(UPLC-QTOF/MS)鑒定蕨麻中化學(xué)成分。方法 Acquity UPLC HSS T3色譜柱(100 mm×2.1 mm,1.8μm);流動(dòng)相:0.05%甲酸水溶液–乙腈,梯度洗脫;體積流量:0.3 mL/min;柱溫:40 ℃;進(jìn)樣體積:2 μL。ESI電噴霧離子源正負(fù)離子掃描,二級(jí)IDA掃描模式下采集碎片數(shù)據(jù)。通過(guò)Sciex OS 2.0數(shù)據(jù)處理軟件結(jié)合數(shù)據(jù)庫(kù)、對(duì)照品、部分文獻(xiàn)和天然產(chǎn)物裂解規(guī)律對(duì)蕨麻水提液中化學(xué)成分進(jìn)行鑒定。結(jié)果 共鑒定出53種化學(xué)成分,其中氨基酸類10個(gè),酚酸類8個(gè),萜酸類4個(gè),黃酮類7個(gè),季胺類生物堿1個(gè),其他類23個(gè)。以野鴉椿酸、鞣花酸為例進(jìn)行碎片離子裂解推測(cè)。結(jié)論 該方法準(zhǔn)確穩(wěn)定,適用于蕨麻中化學(xué)成分的鑒定,為后續(xù)的質(zhì)量控制和物質(zhì)基礎(chǔ)研究提供參考。

[Abstract]
Objective To identify the chemical constituents in Potentillae Radix by UPLC-QTOF/MS. Methods The separation was carried out on Acquity UPLC HSS T3 column (100 mm × 2.1 mm, 1.8 μm) with the mobile phase of 0.05% formic acid - acetonitrile by gradient elution. The flow rate was 0.3 mL/min, column temperature was 40 ℃, and the volume of sample injection was 2 μL. Electrospray ionization (ESI) was used. Positive and negative ion scanning of ESI electric spray ion source was used, and fragmented data were collected under the second level IDA scanning mode. Identification of chemical components in Potentillae Radix water extract were speculated using Sciex OS 2.0 data processing software combined with databases, reference substances, some literature, and natural product cleavage pathways. Results A total of 53 chemical constituents were identified, including 10 amino acids, 8 phenolic acids, 4 terpene acids, 7 flavonoids, 1 quaternary amine alkaloid, and 23 others. The fragmentation process of fragment ions was speculated taking gallic acid and tannic acid as example. Conclusion The method is accurate, stable, and suitable for identification of chemical constituents in Potentillae Radix, and provides reference for subsequent quality control and material basis research.
