50 為 134.87 μg/mL。結(jié)果發(fā)現(xiàn)藻紅蛋白作用 72 h 后,細(xì)胞呈現(xiàn)變大變扁平,細(xì)胞內(nèi)形成空泡、顆粒增多等明顯衰老形態(tài)。在倒置顯微鏡下觀察,酸性 β-半乳糖苷酶染色呈陽性;在熒光顯微鏡下觀察,DAPI 染色出現(xiàn)衰老相關(guān)異染色質(zhì)聚集現(xiàn)象。結(jié)論:藻紅蛋白可通過誘導(dǎo) MCF-7 細(xì)胞衰老達(dá)到抗腫瘤作用。;Objective: To investigate the effects of phycoerythrin(PE)on the induction of cell senescence in human breast cancer MCF-7 cells. Methods: The inhibitory rate of MCF-7 cells growth was measured by MTT assay and morphological changes of MCF-7 cells was observed through the inverted microscope. Acidic β-galactosidase dying (SA-β-Gal dying) and senescence-associated heterochromatin aggregation were used to detect the cell senescence caused by PE in MCF-7 cells. Results: The results of MTT assay indicates that PE could inhibit the growth of human breast cancer MCF-7 cells significantly and the IC50 was 134.87 μg/mL. After 72 h, MCF-7 cells which were treated by PE had become bigger and flatter meanwhile the appearance of vacuolization and particles which are the specific patterns of the cell senescence were also observed. The staining of acidic β-galactosidase was positive, which observed by inverted microscope, and, the emergence of senescence-associated heterochromatin aggregation (SAHF) in staining of DAPI was found under the fluorescence microscope.Conclusion: The results demonstrate that PE has the antitumor effect by inducing the cell senescence of MCF-7 cells."/>
