[摘要]
目的:利用基因表達(dá)譜芯片探索甘草酸協(xié)同麻黃堿對(duì)人支氣管平滑肌細(xì)胞的協(xié)同作用機(jī)制。方法:采用TNF-α誘導(dǎo)人支氣管平滑肌細(xì)胞建立炎癥模型,再分別用麻黃堿、麻黃堿聯(lián)合甘草酸進(jìn)行干預(yù)。提取細(xì)胞的mRNA,反轉(zhuǎn)錄后與基因表達(dá)譜芯片進(jìn)行雜交,經(jīng)掃描分析篩選表達(dá)差異的基因。 結(jié)果:共篩選出顯著表達(dá)差異基因53種,其中33 種基因表達(dá)水平上調(diào),20 種基因表達(dá)下調(diào),經(jīng)Gene Go 軟件分析這些基因在不同生物途徑中存在協(xié)同作用。結(jié)論:甘草酸在G 蛋白偶聯(lián)受體途徑、細(xì)胞周期與凋亡以及離子通道等不同途徑協(xié)同麻黃堿發(fā)揮作用。

[Abstract]
Objective: To investigate the synergic effect of glycyrrhizic acid and ephedrine on the human bronchial smooth muscle cells with cDNA microarray method. Methods: After establishment of inflammation model induced by TNF-α, the human bronchial smooth muscle cells were divided into three groups randomly: the model group, ephedrine group, and glycyrrhizic acid plus ephedrine group. Total RNA of the subcutaneous tumors was extracted using Trizol reagents, and cDNA-labeled fluorescence was synthesized by reverse transcription method. Total DNA preparations from the cells were hybridized to the gene chips. Then the array was scanned and the signals were analyzed after hybridization. Results: Compared with ephedrine group, 33 genes were up-regulated by more than two folds and 20 genes were down-regulated in the HBAMC cells treated with glycyrrhizic acid plus ephedrine. The Gene Go analysis revealed that these genes were associated with G-Protein coupled receptor signaling pathway, cell cycle, apoptosis, and so on. Conclusion: The results show that these pathways may be responsible for the synergic effect of glycyrrhizic acid and ephedrine.
