[摘要]
目的:通過觀察大鼠心肌缺血后,芪參益氣滴丸對其缺血區(qū)組織中鈣調(diào)神經(jīng)磷酸酶B亞基(Ppp3R1)和ATP合成酶亞基δ(Atp5d)基因表達的影響,探討芪參益氣滴丸對急性心肌缺血的保護作用機制。方法:24只大鼠隨機分為正常對照組、模型組、芪參益氣滴丸組,結(jié)扎左冠狀動脈前降支主干建立大鼠心肌缺血模型。采用實時熒光定量PCR(real-time PCR)法檢測不同組間Ppp3r1 和Atp5d基因表達變化。結(jié)果:與空白對照組比較,模型組Ppp3r1基因表達量明顯升高(P<0.01),Atp5d基因表達量明顯降低(P<0.05)。與模型組比較,芪參益氣滴丸組Ppp3r1基因表達量降低(P<0.05),Atp5d基因表達量明顯升高(P<0.01)比較差異均具有統(tǒng)計學(xué)意義。結(jié)論:芪參益氣滴丸對急性心肌缺血的作用保護機制可能與其能夠降低Ppp3r1基因表達量,同時調(diào)控Atp5d基因升高有關(guān)。

Objective: To investigate the protective mechanism of Qishenyiqi Pill in acute myocardial ischemia of rats by observing that Qishenyiqi Pill affected the gene expression of gene Ppp3r1 and Atp5d in myocardial ischemia of rats. Methods: Twenty four rats were randomly divided into normal control group, model group, and Qishenyiqi Pill therapeutic group and then the left anterior descending coronary artery (LAD) was ligated to establish the cardiac ischemia model in rats. The respective group differences in Ppp3r1 and Atp5d was detected by real-time fluorescence quantitative PCR. Results: Compared with control group, Ppp3r1 gene expression in model group was increased significantly (P<0.01), while Atp5d decreased (P<0.05).Compared with model group, Ppp3r1 gene expression in Qishenyiqi Pill therapeutic group was decreased significantly (P<0.05), while Atp5d was increased (P<0.01). Conclusion: The protection mechanism of Qishenyiqi Pill in acute myocardial ischemia may be related to the regulation of gene expression in Ppp3r1 and Atp5d..
