[摘要]
目的:研究治咳川貝枇杷滴丸對(duì)豚鼠的鎮(zhèn)咳、平喘作用。方法:采用豚鼠離體氣管平滑肌收縮實(shí)驗(yàn)和組胺引喘的活體動(dòng)物實(shí)驗(yàn)對(duì)治咳川貝枇杷滴丸的平喘作用進(jìn)行評(píng)價(jià)。在離體實(shí)驗(yàn)中測(cè)定藥物對(duì)乙酰膽堿引起的氣管平滑肌收縮的收縮曲線,計(jì)算收縮百分率;在整體動(dòng)物實(shí)驗(yàn)中測(cè)定給藥后豚鼠的延喘時(shí)間用來表征藥物平喘作用的強(qiáng)弱。同時(shí)采用枸櫞酸誘發(fā)豚鼠咳嗽模型,通過記錄引咳后5 min內(nèi)的咳嗽次數(shù),觀察治咳川貝枇杷滴丸的鎮(zhèn)咳作用。結(jié)果:治咳川貝枇杷滴丸能有效減少豚鼠的咳嗽次數(shù),顯著延長(zhǎng)咳嗽潛伏期;對(duì)豚鼠離體氣管收縮的抑制效果較好;對(duì)組胺致喘豚鼠的哮喘潛伏期有明顯的延長(zhǎng)作用。結(jié)論:治咳川貝枇杷滴丸具有良好的鎮(zhèn)咳、平喘藥理活性,為藥物的臨床應(yīng)用和進(jìn)一步開發(fā)提供了依據(jù)。

[Abstract]
Objective: To study antitussive and anti-asthmatic effect of Zhike Chuanbei Pipa Diwan (ZCPD) in guinea pigs. Methods: Guinea pig models of isolated trachea in vitro and hitamine-induced asthma in vivo were used to investigate the anti-asthmatic effect of ZCPD. In vitro experiment, concentration-response curves of contractile isolated trachea induced by acetylcholine (Ach) were obtained and the concentration producing 50% of the maximal response was determined. In vivo experiment, the latent period of animals after administration was uesd to demonstrate the anti-asthmatic effect. Meanwhile, the aminal cough model induced by citric acid was developed to evaluate the antitussive effect. Qualified guinea pigs were divided into five groups randomly: high, medium, low concentration ZCPD groups, pentoxyverine positive control group, and control group. The number of cough during three minutes was recorded after the cough was induced. Results: ZCPD decreased the frequency of cough and prolonged the latency of induced cough. Morever, it significantly inhibited Ach-induced trachea contractions and prolonged the latent period of asthma by hitamine in guinea pigs. Conclusion: ZCPD has remarkable antitussive and anti-asthmatic effects, which provides the evidence for clinical application and further development.
