[摘要]
目的 觀察金雞菊提取物70%乙醇洗脫物對(duì)高血壓小鼠血壓、血漿血管緊張素Ⅰ(AngⅠ)、血漿血管緊張素Ⅱ(AngⅡ)和組織中AngⅡ濃度的影響。方法 采用高鹽(80 mg/mL NaCl溶液)冷激法,連續(xù)20 d,形成小鼠高血壓模型;將實(shí)驗(yàn)小鼠分為正常組、模型組、卡托普利組(40 mg/kg)和金雞菊提取物70%乙醇洗脫物低、中、高劑量(40、80、160 mg/kg)組。連續(xù)ig給藥28 d后,采用頸動(dòng)脈插管法測(cè)定小鼠血壓并取血,放射免疫法測(cè)定血漿AngⅠ、AngⅡ、及腎組織中AngⅡ的含量。結(jié)果 模型組血漿AngⅠ和組織中AngⅡ含量均明顯高于正常組(P<0.05),其血壓和血漿AngⅡ含量顯著高于正常組(P<0.01或P<0.001);與模型組相比,金雞菊提取物70%乙醇洗脫物中、高劑量(80、160 mg/kg)組血漿AngⅠ含量明顯升高(P<0.05),血漿AngⅡ含量極顯著降低(P<0.001),腎組織中的AngⅡ含量顯著降低(P<0.01)。結(jié)論 金雞菊提取物70%乙醇洗脫物可明顯升高AngⅠ水平,減少AngⅡ生成,推測(cè)其降壓機(jī)制主要是通過影響腎素–血管緊張素系統(tǒng),發(fā)揮AngⅠ拮抗劑或血管緊張素轉(zhuǎn)化酶抑制劑的作用。

[Abstract]
Objective To observe the effect of 70% ethanol elution of Coreopsis tinctoria extract (EECTE) on blood pressure, the concentration of angiotensinⅠ(Ang I) and angiotensinⅡ(Ang II) in plasma, and the concentration of AngⅡ in tissue of hypertensive mice. Methods The hypertension model of mice was established by ig giving high-salt water and keeping them in low-temperature environment for 4 h daily for 20 d; The mice were divided into control, model, Captopril (40 mg/kg), low-, mid-, and high-dose (40, 80, and 160 mg/kg) EECTE groups. After ig administration for 28 d, the blood pressure was measured, the blood and kidney tissue were collected, and the plasma concentration of AngⅠ, AngⅡ, and tissue homogenate AngⅡwere determined by radioimmunoassay. Results The AngⅠlevel in plasma and AngⅡlevel in tissue homogenate in model group were obviously higher than those in the control group (P < 0.05), and the blood pressure and concentration of plasma AngⅡ were significantly higher than those in the control group (P < 0.01, 0.001). The mid- and high-dose EECTE increased AngⅠand reduced AngⅡin plasma obviously (P < 0.01), and lessened local AngⅡ in tissue homogenate compared with those in the model group (P < 0.001). Conclusion EECTE may increase AngⅠand reduce AngⅡ markedly, so the possible mechanism in the reduction of blood pressure in hypertensive mice is due to its effect on renin-angiotensin system and the effect of AngⅠ antagonist or angiotensin converting enzyme inhibitor.
