[摘要]
目的 對(duì)db/db小鼠進(jìn)行生物學(xué)特性研究,滿足糖尿病研究的需要。方法 在試驗(yàn)周期內(nèi),測(cè)定db/db小鼠的空腹血糖、體質(zhì)量及攝食飲水量,并測(cè)定21周齡模型動(dòng)物的糖耐量、血中胰島素、胰高血糖素、三酰甘油、總膽固醇等指標(biāo)的水平,進(jìn)行胰腺的免疫組化雙重染色及主要臟器的病理學(xué)檢查,并與db/m小鼠進(jìn)行比較。結(jié)果 與對(duì)照組db/m小鼠比較,db/db小鼠過度肥胖,伴有高血糖、胰島素抵抗、脂質(zhì)代謝紊亂,且肝臟和胰腺組織均出現(xiàn)明顯病變,胰腺免疫組化雙重染色結(jié)果與血液學(xué)測(cè)定結(jié)果一致,并可觀察到胰島中A細(xì)胞和B細(xì)胞的分布變化情況。結(jié)論 對(duì)自發(fā)性2型糖尿病模型db/db小鼠所做的相關(guān)生物學(xué)特性檢測(cè),可供糖尿病研究參考。

[Abstract]
Objective To meet the need of study on diabetes mellitus, we investigated the biological characteristics of db/db mice. Methods In this experiment, fasting blood glucose, body weight, food intake, and water intake were measured. Fasting blood glucose levels in oral glucose tolerance test were observed on db/db mice of 21 weeks old. Meanwhile, the mice were sacrificed for the detection of insulin, glucagon, triglyceride (TG), and total clolesterol (TC) in serum, immunohistochemical double staining in pancreas, and morphological study. All the data of db/db mice were compared with those of db/m mice in statistics. Results Compared with db/m mice, obesity, hyperinsulinemia, hyperglycemia, and lipid metabolism disorder were serious on db/db mice, following obvious pathological deterioration of liver and pancreas. In accordance with the result of blood testing, change in distribution of A cells and B cells could be observed in immunohistochemical double staining on pancreas of db/db mice. Conclusion The db/db mice could be used as appropriate disease models of type 2 diabetes mellitus, providing the relevant biological information for further study in this field.
