[摘要]
目的 通過實驗室前期建立的中藥寒熱藥性的細(xì)胞評價方法評價中藥白頭翁的寒熱藥性。方法 用噻唑藍(lán)(MTT)比色法考察白頭翁對HeLa細(xì)胞和SGC-7901細(xì)胞體外增殖的影響,倒置顯微鏡觀察白頭翁對細(xì)胞形態(tài)特征的影響,臺盼藍(lán)染色法分析白頭翁的細(xì)胞毒作用。結(jié)果 白頭翁在所選質(zhì)量濃度10~400 μg/mL內(nèi)抑制細(xì)胞增殖。形態(tài)學(xué)觀察表明白頭翁能使細(xì)胞密度減小,細(xì)胞固縮變圓,顆粒感增加。臺盼藍(lán)染色表明白頭翁對這兩種細(xì)胞沒有細(xì)胞毒作用。結(jié)論 白頭翁性寒,對所用細(xì)胞沒有細(xì)胞毒作用。

[Abstract]
Objective To evaluate the cold-heat characteristics of Pulsatillae Radix by the method we have set up. Methods MTT assay was used to investigate the effect of Pulsatillae Radix on the proliferation of HeLa and SGC-7901 cells in vitro. The morphological changes of HeLa and SGC-7901 cells treated with Pulsatillae Radix were observed through inverted microscope. Trypan-blue staining was used to analyze the cytotoxicity of Pulsatillae Radix. Results Pulsatillae Radix could inhibit the proliferation of HeLa and SGC-7901 cells in the concentration range of 10—400 μg/mL. Morphological observation showed Pulsatillae Radix in the concentration range we have chosen could decrease the density of HeLa and SGC-7901 cells and condense the cells. Trypan-blue staining showed Pulsatillae Radix had no cytotoxicity on HeLa and SGC-7901 cells. Conclusion Pulsatillae Radix is a cold drug and has no cytotoxicity.
