[摘要]
目的 探討延齡草苷(trillin)對小膠質細胞(BV-2)的炎性激活的抑制作用。方法 不同濃度的延齡草苷孵育0.5 h,然后用脂多糖進行刺激,培養(yǎng)24 h后收集上清,Greiss法檢測NO濃度;CCK-8檢測延齡草苷對小膠質細胞活力的影響;Elisa檢測腫瘤壞死因子(TNF-α)的濃度;實時定量PCR(RT-PCR)檢測TNF-α、IL-1β、IL-6、iNOS、COX-1、COX-2基因的表達。結果 在不影響細胞活力的前提下,延齡草苷能明顯抑制NO的釋放,同時能抑制激活誘導細胞死亡,能明顯抑制TNF-α的產生,能明顯抑制TNF-α、IL-1β、IL-6、iNOS、COX-2 mRNA的表達,但是不能抑制COX-1 mRNA的表達。結論 延齡草苷可以抑制LPS誘導的小膠質細胞的炎性激活,抑制炎性基因的表達。

Objective To study the inhibition of trillin on the inflammatory activation of microglia (BV-2). Methods The BV-2 cells were incubated with trillin at different concentration for 0.5 h and stimulated with lipopolysaccharide (LPS). The supernatant was collected after 24 h culture, and the NO level was detected by Greiss method; The activity of BV-2 cells was detected with CCK-8; The expression of tumor necrosis factor (TNF-α), IL-1β, IL-6, iNOS, COX-1, and COX-2 was detected by real time quantitative (RT-PCR). Results Trillin can inhibit the NO release in microglia induced by LPS without affecting the cell viability and inhibit the inflammatory factors such as TNF-α, IL-1β, IL-6, iNOS, and COX-2. Conclusion Trillin can inhibit the inflammatory activationof BV-2 cells induced by LPS and inhibit the expression of inflammatory factor.
