用于冠心舒膠囊在大鼠體內(nèi)的藥動(dòng)學(xué)。方法 色譜柱為C18柱(50 mm×2.1 mm,1.7 μm),采用梯度洗脫的方式分離被測(cè)物質(zhì),利用液液萃取的方法處理血漿樣品,以多反應(yīng)監(jiān)測(cè)(multiple reaction monitoring,MRM)掃描方式檢測(cè)。結(jié)果 丹參素在線性范圍內(nèi)線性關(guān)系良好,日內(nèi)日間精密度(RSD)均小于15%,準(zhǔn)確度(RE)在±15%以內(nèi),平均提取回收率在80%以上,基質(zhì)效應(yīng)在90%~110%。結(jié)論 該方法適合于冠心舒膠囊在大鼠體內(nèi)的藥動(dòng)學(xué)研究。;Objective To establish an LC- MS/MS method for determing danshensu (DSS) in rat plasma, and to apply the method to the pharmacokinetics of Guanxinshu Capsule in rats. Methods The analytes were separated on a C18 column (50 mm × 2.1 mm, 1.7 μm) and a triple-quadrupole mass spectrometry equipped with electrospray ionization (ESI) source was applied for detection. The liquid-liquid extraction cartridge was employed to extract the analyte from rat plasma, and multiple reaction monitoring was used to detect the samples. Results The method was linear over the concentration ranges of 1—1 000 ng/mL for DSS. The intra- and inter-day relative standard deviation (RSD) were less than 15% and the relative errors were all within 15%, with the recovery of DSS (> 80%) and matrix effect of 90%—110%. Conclusion The method is successfully applied to the pharmacokinetic study of Guanxinshu Capsule in rat."/>
