[摘要]
體內(nèi)彗星試驗(yàn)是采用單細(xì)胞凝膠電泳的方法檢測(cè)體內(nèi)DNA損傷的技術(shù)。具有靈敏度高、操作簡(jiǎn)便、經(jīng)濟(jì)快速等優(yōu)點(diǎn)。隨著遺傳毒性研究的發(fā)展,體內(nèi)彗星試驗(yàn)已經(jīng)成為重要的藥物遺傳毒性評(píng)價(jià)方法。對(duì)動(dòng)物試驗(yàn)階段和彗星試驗(yàn)階段各操作步驟進(jìn)行了歸納總結(jié),以期對(duì)相關(guān)方法學(xué)建立提供參考,并提出采用簡(jiǎn)化和標(biāo)準(zhǔn)化的研究方法將有利于體內(nèi)彗星試驗(yàn)在藥物遺傳毒性評(píng)價(jià)的應(yīng)用。

[Abstract]
The in vivo comet assay is a technology using single cell gel electrophoresis for the detection of DNA damage in vivo. The assay has the advantages of high sensitivity, ease of performance, low cost, and short time. With the development of the genotoxicity research, the in vivo comet assay has become an important method for genotoxicity assessment. This article reviewed some operating procedures in the animal test stage and the comet test stage, and suggested that using simplified and standardized research methods should be good for the application of in vivo comet assay on drug genotoxicity evaluation.
