目的 研究連翹苷元對牛血清白蛋白誘導(dǎo)的大鼠免疫性肝纖維化的影響。方法 動物按照體質(zhì)量隨機(jī)分為對照組、模型組、連翹苷元低、中、高劑量(0.1、0.3、1.0 mg/kg)組和水飛薊賓葡甲胺組(50 mg/kg), 利用牛血清白蛋白建立大鼠免疫性肝纖維化模型。全自動生化分析儀檢測血清蛋白水平;Elisa試劑盒檢測血清透明質(zhì)酸(HA)、層黏蛋白(LN)、Ⅳ型膠原(Ⅳ-C)和Ⅲ型前膠原(PCⅢ)水平;分光光度計(jì)法檢測肝臟組織羥脯氨酸(Hyp);取肝左葉固定、HE染色, 顯微鏡下觀察肝纖維化程度。結(jié)果 連翹苷元能夠顯著降低肝纖維化大鼠血清HA、LN、Ⅳ-C和PCⅢ的量, 減少肝臟質(zhì)量和肝臟系數(shù), 降低肝臟Hyp水平, 減輕肝纖維化程度。結(jié)論 連翹苷元對免疫性大鼠肝纖維化有較好的治療作用。

[Abstract]
Objective To investigate the effects of forsythiaside on immunological hepatic fibrosis induced by bovine serum albumin (BSA) in rats. Methods To establish the immunological hepatic fibrosis models induced by BSA. Animals were randomly divided into six groups namely normal group, model group, forsythiaside (0.1, 0.3, and 1.0 mg/kg) treated groups, and Silybin group (50 mg/kg). The levels of TP, ALB, AST, and ALT were assayed by automatic biochemical analyzer; The levels of LN, HA, Ⅳ-C and PCⅢ in serum were assayed by ELISA kit; Hydroxyproline (Hyp) content in liver tissue was assayed by spectrophotometry; Taking the left hepatic lobe fixed, HE staining was used to examine the histopathological change under microscope. Results Forsythiaside could significantly alleviate the hepatic fibrosis, not only in decreasing the Hyp content in hepatic tissue, but also reducing the levels of LN, HA, Ⅳ-C and PCⅢ in rats with immunological hepatic fibrosis. Conclusion Forsythiaside has the protective effects on the hepatic fibrosis induced by BSA in rats.
