[摘要]
目的 觀察潰結(jié)康經(jīng)灌腸給藥對(duì)三硝基苯磺酸(trinitrobenzene sulfonic acid,TNBS)誘導(dǎo)的炎癥性腸病動(dòng)物模型的作用,以明確潰結(jié)康對(duì)炎癥性腸病的治療作用,為探索藥物應(yīng)用于克羅恩病治療的可行性。方法 采用TNBS灌腸的方法誘導(dǎo)大鼠潰瘍性結(jié)腸炎模型,觀察潰結(jié)康灌腸給藥對(duì)大鼠疾病活動(dòng)指數(shù)(DAI)評(píng)分、血清干擾素-γ(IFN-γ)、白介素-l(IL-1)、白介素-4(IL-4)、白介素-10(IL-10)和腸黏膜中谷氨酰胺(Glutamine,Gln)的含量的影響,并觀察藥物對(duì)大鼠結(jié)腸組織損傷評(píng)分以及病理學(xué)的影響。結(jié)果 TNBS誘導(dǎo)的結(jié)腸炎模型可引起動(dòng)物體質(zhì)量下降,肉眼血便或便潛血陽性,DAI評(píng)分增高,血清IFN-γ,IL-1升高,腸黏膜Gln降低,潰結(jié)康不同劑量組灌腸給藥可改善TNBS引起的DAI評(píng)分增高以及降低血清IFN-γ、IL-1水平,升高腸黏膜Gln,緩解結(jié)腸炎組織學(xué)變化,其中尤以潰結(jié)康高劑量組作用更為明顯。結(jié)論 潰結(jié)康灌腸給藥對(duì)緩解TNBS誘導(dǎo)結(jié)腸炎模型有一定治療作用。

[Abstract]
Objective To observe the effect of Kuijiekang on model induced by trinitrobenzene sulfonic acid, to examine its efficacy for inflammatory bowel disease, and to explore the feasibility of this medicine used for treatment of Cronh's disease. Methods Rat model of inflammatory bowel disease was set up by rectally administration of TNBS, and the effects of Kuijikang on indexes of DAI, IFN-γ, IL-1, and IL-4, as well as and the content of glutamine (Gln) in rats were determined. Results The rats with inflammatory bowel disease induced by TNBS have decreased body weight, macroscopical or microscopical hematochezia, increased DAI score, increased levels of IFN-γ and IL-1, and decreased Gln. Different doses of Kuijiekang could significantly improve the change of DAI score, lower the levels of IFN-γ and IL-1, raise the Gln in intestinal mucosa caused by TNBS, and relieve the histology change of colon inflammation, the highest dose with the most significant effect. Conclusion Kuijiekang rectlly administerd is effective for the colitis model induced by TNBS.
