[摘要]
目的 用莫西沙星、特非那定和卡托普利對(duì)犬植入式生物遙測(cè)技術(shù)進(jìn)行性能評(píng)價(jià)研究。方法 4只埋植有遙測(cè)植入子的清醒Beagle犬,通過(guò)拉丁方設(shè)計(jì),在4個(gè)不同的給藥日分別ig給予空白膠囊、莫西沙星、特非那定和卡托普利30 mg/kg,用DSI遙測(cè)系統(tǒng)連續(xù)采集給藥前2 h至給藥后24 h的生理信號(hào),對(duì)心電、血壓、體溫等數(shù)據(jù)進(jìn)行分析,比較給藥前與給藥后各時(shí)間點(diǎn)各指標(biāo)的差異。結(jié)果 空白膠囊對(duì)犬的各項(xiàng)生理指標(biāo)無(wú)明顯影響;給予莫西沙星和特非那定后,犬QT間期及校正的QT間期(QTc)與給藥前比較,出現(xiàn)不同程度的延長(zhǎng);給予卡托普利對(duì)犬的心電參數(shù)無(wú)影響但顯著降低血壓。結(jié)論 植入式生物遙測(cè)技術(shù)可靈敏的檢測(cè)到藥物對(duì)清醒犬心血管系統(tǒng)的影響,可用于安全藥理研究和清醒犬心血管模型研究。

[Abstract]
Objective To evaluate the canine telemetry assay using moxifloxacin, terfenadine, and captopril. Methods Four conscious Beagle's dogs were instrumented with implantable telemetry devices for the measurement of blood pressure, ECG, and body temperature. Dogs were administered with blank capsules, moxifloxacin, terfenadine, and captopril in four different days using a 4×4 Latin's square design. The cardiovascular parameters were collected from 2 h before dosing to 24 h post-dosing. Results Blank capsules showed no significant effect on cardiovascular parameters. Moxifloxacin and terfenadine induced different prolongation on QT interval and corrected QT interval (QTc). Captopril had no effect on the ECG but reduced blood pressure significantly. Conclusion Telemetry assay is recommended as a sensitive test system for safety pharmacology evaluation and canine cardiovascular model which can detect the impact of drugs on the cardiovascular system.
