[摘要]
目的 通過UFLC-QTOF-MS技術,評價通脈顆粒的質量。方法 對10個生產(chǎn)企業(yè)的61批次的通脈顆粒,采用Agilent proshell 120(100 mm×3.0 mm,2.7 μm),流動相是甲醇-緩沖鹽,梯度洗脫,體積流量0.4 mL/min,柱溫40℃,ESI源,正離子模式下采集質譜數(shù)據(jù)。應用Markerview軟件進行主成分分析(PCA)對通脈顆粒進行分組。結果 正離子模式時PCA分析能很好的區(qū)分通脈顆粒的質量,通過t檢驗找出了大豆素8-C-芹糖(1,6)-O-葡萄糖苷和1,5-Anhydro-1-[5,7- dimethoxy-2-(4-methoxyphenyl)-4-oxo-4H-chromen-8-yl]hexitol兩個差異最大的化合物。結論 PCA分析可以作為評價通脈顆粒質量的分類方法。

[Abstract]
Objective To evaluate the quality of Tongmai Granule by UFLC-QTOF-MS. Methods The UFLC was performed on Agilent proshell 120 column (100 mm×3.0 mm, 2.7 μm) with the mobile phase of MeOH-buffer salt (43:57) by gradient elution, the flow rate was 0.4 mL/min, the column temperature was 40℃, electrospray ionization source was used under the positive ion mode. Tongmai Granule was classified by principle component analysis (PCA) with Markerview software. Results PCA analysis could distinguish the quality of Tongmai Granule under positive ion mode. Daidzein 8-C-apiosyl(1,6)-O-glucoside and 1,5-anhydro-1-[5,7-dimethoxy-2-(4-methoxyphenyl)-4-oxo-4H-chromen-8-yl]hexitol were found to be the biggest different compounds by t test. Conclusion PCA is an effective classification method to evaluate the quality of Tongmai Granule.
