[摘要]
目的 對木香莖葉提取物化學(xué)成分進(jìn)行初步檢識,研究其對小鼠腸推進(jìn)和胃排空的影響。方法 制備木香莖葉水、乙醇、乙醚提取物,化學(xué)反應(yīng)檢識木香莖葉化學(xué)成分;以小鼠為研究對象,新斯的明和阿托品誘導(dǎo)小鼠胃腸亢進(jìn)和抑制狀態(tài),ig給予0.5 g/kg木香莖葉水提物、醇提物,用改良的酚紅法測定其對胃腸正常、亢進(jìn)、抑制狀態(tài)小鼠胃排空率和小腸推進(jìn)率的影響。結(jié)果 木香莖葉醇提物、水提物均能夠顯著提高正常小鼠小腸酚紅推進(jìn)率(P<0.05、0.01),顯著降低新斯的明所致胃腸亢進(jìn)狀態(tài)下的小鼠小腸酚紅推進(jìn)率(P<0.05),且醇提物的作用較強(qiáng);兩提取物使阿托品所致抑制狀態(tài)下小鼠小腸推進(jìn)率進(jìn)一步降低,且差異顯著(P<0.05),兩提取物之間無明顯差異;兩種提取物對三種狀態(tài)下的胃排空都發(fā)揮顯著抑制作用(P<0.05、0.01)。化學(xué)成分檢識表明,木香莖葉中含蛋白、糖、揮發(fā)油、黃酮、內(nèi)酯、皂苷、生物堿和鞣質(zhì)等多種成分,但何種成分與小腸推進(jìn)和胃排空有關(guān)尚不確定。結(jié)論 木香莖葉提取物對正常小鼠的小腸推進(jìn)功能具有一定的促進(jìn)作用,對新斯的明所致小腸推進(jìn)亢進(jìn)有抑制作用,對阿托品所致小腸推進(jìn)抑制有加劇作用,其對腸道的促進(jìn)作用可能與M膽堿受體有關(guān);對正常和亢進(jìn)小鼠的胃排空有抑制作用,對阿托品所致胃排空抑制有加劇作用。

[Abstract]
Objective To investigate the constituents of water and ethanol extracts from the stems and leaves of Aucklandia lappa and their effect on gastric emptying and intestinal propulsion of gastrointestinal normal, hyperthyroidism and inhibited mice. Methods The water, ethanol and aether petrolei extracts from stems and leaves of A. lappa were prepared, and the chemical constituents of the stems and leaves were identified by chemical reaction. The hyperthyroidism state was induced by neostigmine and inhibiting state was induced by atropine. Mice were ig administered with water and ethanol extracts (0.5 g/kg), with the improved phenol red method to determine gastric emptying and intestinal propulsion in gastrointestinal normal, hyperthyroidism and inhibited mice. Results Both water and ethanol extracts significantly improved intestinal propulsion in normal mice (P<0.05 and 0.01), while significantly decreased the intestinal propulsion in hyperthyroidism mice (P<0.05) and ethanol extract showed a stronger decreasing effect than that of water extract. The inhibitory effect of atropine on intestinal propulsion was intensified by these two extracts (P<0.05). The restrained gastric emptying of normal, hyperthyroidism and inhibited mice was also observed. Chemical composition analysis indicated that many kinds of chemical components including protein, sugar, essential oil, flavonoids, lactones, alkaloids, saponins and tannins existed in the leaves and stems of A. lappa. Conclusion The leaves and stems of A. lappa could promote the intestinal propulsion of normal mice and restrain the intestinal propulsion of hyperthyroidism and inhibited mice while inhibit the gastric emptying in any condition tested, and its promoting effect on the gut may be related to the M cholinergic receptor.
