[摘要]
目的 體內(nèi)考察鼻咽清顆粒對(duì)人鼻咽癌細(xì)胞(CNE-2)裸鼠種植瘤的放療增敏作用并對(duì)其可能機(jī)制進(jìn)行初步探索。方法 建立CNE-2細(xì)胞的裸鼠種植瘤模型,取荷瘤成功裸鼠30只,隨機(jī)分成5組,分別為模型組,鼻咽清顆粒低、高劑量(生藥劑量5.2、10.4 g/kg)組,放療組,鼻咽清顆粒(生藥劑量5.2 g/kg)+放療組,每天2次ig給藥;放療照射劑量為4 GY,隔日1次,連續(xù)2周。給藥期間每5天測(cè)量腫瘤體積,至給藥30 d處死,取瘤組織,稱質(zhì)量并計(jì)算抑瘤率;免疫組化檢測(cè)P53、Bcl-2蛋白表達(dá)。結(jié)果 與模型組比較,各治療組均對(duì)移植瘤體積、質(zhì)量發(fā)揮顯著抑制作用(P<0.05、0.01);鼻咽清顆粒+放療組瘤體積、質(zhì)量均顯著低于放療組(P<0.05、0.01)。免疫組化結(jié)果提示,與模型組比較,各治療組移植瘤組織中P53、Bcl-2蛋白表達(dá)陽(yáng)性細(xì)胞率均顯著降低(P<0.05、0.01);與放療組比較,鼻咽清顆粒+放療組P53、Bcl-2蛋白表達(dá)陽(yáng)性細(xì)胞率顯著降低(P<0.01)。結(jié)論 鼻咽清顆粒抑制鼻咽癌的生長(zhǎng)且具有放療增敏作用,可能與同時(shí)抑制突變性P53和Bcl-2蛋白表達(dá)相關(guān)。

[Abstract]
Objectives To study the radiosensitization effect of Bi-Yan-Qing granules on xenograft tumor of human nasopharyngeal carcinoma cell line CNE-2 in the BALB/C nude mice and explore the possible mechanism. Methods CNE2 cells were injected subcutaneously into nude mice to establish transplanted tumor mice model, Then, 30 mice were randomly divided into five groups:model group, Bi-Yan-Qing Granules low and high dosage (crude drug dosage:5.2, 10.4 g/kg) groups, irradiation group, and Bi-Yan-Qing Granules (crude drug dosage:5.2 g/kg) combined with irradiation group. Ig administered for twice a day, radiotherapy dose was 4 GY, once two days for two weeks. The tumor volume was measured every five days during dosing period. Nude mice were killed after 30 days administration, and the tumor tissue was weighed to calculate tumor inhibition rate. The immunohistochemistry assay was used to detect the expression levels of P53 and Bcl-2 protein in tumor tissues in each group. Results Compared with model group, the transplantation tumor volume and weight of each treatment group were significantly inhibited (P < 0.05, 0.01). The tumor volume and quality of combination group (Bi-Yan-Qing Granules and irradiation) were significantly lower than those of the radiotherapy group (P < 0.05, 0.01). Immunohistochemistry results showed that, compared with model group, the positive cellular rate of P53 and Bcl-2 protein expression were significantly decreased in tumor tissue of each treatment group (P < 0.05, 0.01). Compared with radiotherapy group, the positive cellular rate of P53 and Bcl-2 protein expression in the combination group was significantly reduced (P < 0.01). Conclusions Bi-Yan-Qing Granules inhibit the growth of nasopharyngeal carcinoma and have a radiosensitization effect, which may be related to the inhibition of mutation in P53 and Bcl-2 protein expression at the same time.
