目的 研究腸炎寧顆粒作用于ICR幼齡小鼠的止瀉抗炎藥效及體外抑菌作用,為其臨床兒童用藥提供參考。方法 以ICR幼齡小鼠為考察對象,ig給予高、中、低劑量(2.500、1.250和0.625 g/kg)腸炎寧顆粒及蒙脫石散(陽性藥,4.2 g/kg),連續(xù)給藥3 d后,采用番瀉葉浸出物和蓖麻油致瀉模型、腸推進(jìn)實(shí)驗(yàn)、冰醋酸致腹腔毛細(xì)血管通透性增加模型考察腸炎寧顆粒的止瀉、抗炎作用;體外培養(yǎng)表皮葡萄球菌和大腸埃希菌,加入571.00、285.50、142.75、71.38、35.69、17.84、8.92、4.46、2.23和1.12 mg/mL的腸炎寧顆粒,觀察其抑菌作用。結(jié)果 與模型組比較,0.625、1.250和2.500 g/kg的腸炎寧顆??梢燥@著減少番瀉葉浸出物、蓖麻油作用0~5 h內(nèi)的腹瀉樣便數(shù)(P<0.05、0.001);與對照組比較,2.5 g/kg的腸炎寧顆??梢燥@著降低腸推進(jìn)系數(shù)(P<0.05);0.625、1.250和2.500 g/kg的腸炎寧顆??梢燥@著減少冰醋酸所致的幼鼠腹腔毛細(xì)血管通透性增加(P<0.05、0.01)。8.92 mg/mL的腸炎寧顆粒即可在體外抑制表皮葡萄球菌和大腸埃希菌的生長,為最低抑菌濃度。結(jié)論 腸炎寧顆粒具有止瀉、抑制腸胃推進(jìn)及對抗急性滲出性炎癥的作用;體外可以抑制表皮葡萄球菌和大腸埃希菌的生長。

[Abstract]
Objective The purpose of this article is to investigate the anti-diarrheal, anti-inflammatory and bacteriostatic effects of Changyanning granule in weaning mice, and provide reference for its pediatric use. Methods The ICR model of weaning mice was established and administrated with low, middle and high dose (0.625, 1.25, 2.50 g/kg) of Changyanning granuleand smecta (positive control, 4.20 g/kg). After continuous administration for three days, diarrhea models caused by castor oil and Senna Leaf extract, intestinal propulsion test and abdominal capillary permeability increase model caused by glacial acetic acid were used to test the antidiarrheal and anti-inflammatory effects of Changyanning granule. In vitro experiment was also performed to test the bacteriostatic effect of Changyanning granule. Staphylococcus epidermidis and Escherichia coli were cultured and treated with different concentrations of Changyanning granule (571.00, 285.50, 142.75, 71.38, 35.69, 17.84, 8.92, 4.46, 2.23, and 1.12 mg/mL). Results Compared with model group, diarrhea stool numbers were significantly reduced (P < 0.05, 0.001) after the dosing of Senna Leaf extract and castor oil within 5 h in 0.625, 1.25 and 2.50 g/kg Changyanning granule groups. Intestinal propulsion coefficient was reduced significantly (P < 0.05) in 2.50 g/kg Changyanning granule group in comparison with control group; the inhibitory actions of Changyanning (0.625, 1.25, 2.50 g/kg) on the increase of glacial acetic acid-induced abdominal capillary permeability were obviously (P < 0.05, 0.01); The growth of Staphylococcus epidermidis and Escherichia coli were inhibited with the initial concentration of 8.92 mg/mL in vitro. Conclusion Changyanning granule showed anti-diarrheal and acute exudative inflammation inhibition effects in weaning mice, and it could also inhibit the growth of Staphylococcus epidermidis and Escherichia coli in vitro.
