[摘要]
目的 基于中藥多成分作用特點,探討黃芪多成分在體內(nèi)的代謝情況,遴選黃芪質(zhì)控成分。方法 制備黃芪水提物,ig給予大鼠(生藥20 g/kg),每天3次,連續(xù)給藥3 d,腹主動脈采血、代謝籠收集尿液,結(jié)合超高效液相色譜-串聯(lián)質(zhì)譜(UPLC-MS/MS)技術(shù)對黃芪水提物主要成分、入血成分和尿液排泄成分進行鑒別,并分析主要的代謝途徑,遴選黃芪質(zhì)控成分。結(jié)果 黃芪水提物主成分共鑒別出包括毛蕊異黃酮葡萄糖苷、芒柄花苷、calycosin-7-O-glc-6"-O-malonate、毛蕊異黃酮、芒柄花素等在內(nèi)的15個化合物;血漿樣品中指認(rèn)出11個成分,包括7個代謝成分和4個原型成分;尿液樣品中指認(rèn)出14個成分,9個代謝成分及5個原型成分。結(jié)論 根據(jù)質(zhì)譜數(shù)據(jù)分析結(jié)果,遴選出黃芪中以原型入血的成分和參與體內(nèi)代謝的成分,包括毛蕊異黃酮葡萄糖苷、芒柄花苷、毛蕊異黃酮、芒柄花素、大豆苷元、3-hydro-9,10-diMP、isomer of 3-hydro-9,10-diMP、calycosin-7-O-glc-6"-O-acetate和formononetin-7-O-glc-6"-O-acetate,作為黃芪的質(zhì)控成分。

[Abstract]
Objective To investigate the internal metabolism of Astragali Radixin and to select the quality control components of Astragali Radix by using UPLC-MS/MS based on the feature of multi-component.Methods Water extract of Astragali Radix was prepared for ig administration (crude drug 20 g/kg) to rats for 3 d, three times a day. The metabolic components in the rats blood from abdominal aorta and urine collected by metabolic cage, as well as water extracts after oral administration were analyzed by UPLC-MS/MS with view to analyzing the metabolic pathway and the quality control components of Astragali Radix.Results There are 15 known constituents identified in the decoction of Astragali Radix, including calycosin-7-O-β-D-glucoside, ononin, calycosin-7-O-glc-6"-O-malonate, calycosin, formononetin etc. Eleven constituents were detected in the blood, including seven metabolic components, and four prototypes. There are 14 constituents identified in the Urine sample, including nine metabolic components and five prototypes.Conclusion There 10 components metabolic components and prototypes as quality control ingredients in water extracts of A. Radix based on the results of MS, which included calycosin-7-O-glucoside, ononin, calycosin, formononetin, daidzein, 3-hydro-9,10-diMP, isomer of 3-hydro-9,10-diMP, calycosin-7-O-glc-6"-O-acetate and formononetin-7-O-glc-6"-O-acetate.
