[摘要]
目的 探討固公果根提取物促進(jìn)結(jié)腸癌細(xì)胞凋亡及機(jī)制研究。方法 分別采用MTT法、DAPI染色及流式細(xì)胞術(shù)檢測(cè)固公果根提取物對(duì)結(jié)腸癌細(xì)胞增殖活性及細(xì)胞凋亡的影響;蛋白免疫印跡(Western-Blot)法檢測(cè)凋亡相關(guān)蛋白的活性表達(dá)水平。結(jié)果 固公果根提取物能顯著抑制結(jié)腸癌SW620和LOVO細(xì)胞增殖,促進(jìn)兩種結(jié)腸癌細(xì)胞凋亡,上調(diào)凋亡相關(guān)基因Keap-1、Bax的蛋白表達(dá),下調(diào)Nrf2、HO-1以及Bcl2蛋白的表達(dá)。結(jié)論 固公果根提取物能顯著抑制人結(jié)腸癌細(xì)胞SW620和LOVO增殖、誘導(dǎo)細(xì)胞凋亡,其作用機(jī)制可能涉及調(diào)控凋亡相關(guān)基因Bax、Bcl-2表達(dá),且與激活Nrf2氧化應(yīng)激通路有關(guān)。

[Abstract]
Objective To investigate effects and mechanism of the extract of roots of Rose odorata sweet var. gigantean (ROE) on apoptosis of colon cancer cells. Methods MTT assay, DAPI staining, and flow cytometry were used to detect the effects of ROE extracts on proliferation and apoptosis of colon cancer cells. The protein expression was detected by Western Blotting. Results ROE extracts could significantly inhibit the growth of SW620 and LOVO cells and promote it apoptosis. ROE extracts could also up-regulate the expression of Keap-1 and Bax protein, and down-regulate the expression of Nrf2, HO-1 and Bcl2 protein. Conclusion ROE extracts can significantly inhibit the proliferation and induce apoptosis of human colon cancer cells SW620 and LOVO, which mechanisms might be involved in regulating the expression of apoptosis-related genes Bax and Bcl-2 and activating Nrf2 oxidative stress pathway.
