+CD25+FOXP3+、CD3-CD16+CD56+、CD4+CD45RA+CCR7+的細(xì)胞比例;ELISA法檢測白細(xì)胞介素(IL)-2、IL-4、IL-10、γ-干擾素(INF-γ)細(xì)胞因子分泌情況。結(jié)果 與陰性對照組比較,hUC-MSCs對淋巴細(xì)胞的增殖具有顯著抑制作用(P<0.05),抑制效果隨數(shù)量增加而增強(qiáng);hUC-MSCs能夠顯著增加共培養(yǎng)體系中CD4+CD25+FOXP3+細(xì)胞比例(P<0.05),顯著降低CD4+CD45RA+CCR7+細(xì)胞比例(P<0.05),對CD3-CD16+CD56+細(xì)胞比例無顯著影響;hUC-MSCs能夠顯著降低IL-2、IL-4、INF-γ水平,顯著增加IL-10的分泌水平(P<0.05)。結(jié)論 hUC-MSCs具有免疫抑制功能。;Objective To observe the effects of human umbilical cord mesenchymal stem cells (hUC-MSCs) on the proliferation of allogeneic total lymphocytes and different lymphocyte subsets, as well as the secretion of cytokines. Methods hUC-MSCs and peripheral blood lymphocytes of healthy adults were isolated and cultured; MTT method was used to detect the proliferation of lymphocytes by after co-culture for 3 days; the proportion of CD4+CD25+FOXP3+, CD3-CD16+CD56+ and CD4+CD45RA+CCR7+ in the co-culture system was detected by flow cytometry; ELISA was used to detect the secretion of IL-2, IL-4, IL-10 and INF-γ. Results hUC-MSCs could significant inhibit the proliferation of T lymphocytes, and the anti-proliferative effect was enhanced with the increase of the number of lymphocytes; the proportion of CD4+CD25+FOXP3+ cells in co-culture system was increased by hUCMSCs (P<0.05); the proportion of CD4+CD45RA+CCR7+ cells showed a downward tendency, and the difference was statistically significant (P<0.05); however, there was no significant difference in CD3-CD16+CD56+ cells. hUC-MSCs could reduce the levels of IL-2, IL-4 and INF-γ, but increase the secretion level of IL-10, and the difference was statistically significant (P<0.05).
