[摘要]
目的 基于HPLC指紋圖譜、多成分定量結(jié)合化學(xué)模式識(shí)別法評(píng)價(jià)黔鉤藤質(zhì)量。方法 從貴州中藥材市場(chǎng)隨機(jī)購(gòu)買了18批鉤藤飲片,采用HPLC法建立了18批鉤藤飲片的指紋圖譜;通過(guò)中藥色譜指紋圖譜相似度評(píng)價(jià)系統(tǒng)(2012年版)進(jìn)行相似度評(píng)價(jià),對(duì)其中5種生物堿進(jìn)行了指認(rèn),并采用外標(biāo)法和一測(cè)多評(píng)法進(jìn)行了含量測(cè)定;采用SPSS 26.0統(tǒng)計(jì)軟件進(jìn)行聚類分析和主成分分析。結(jié)果 在鉤藤飲片指紋圖譜中,確定了共有峰28個(gè),采用對(duì)照品指認(rèn)了5個(gè)生物堿成分,分別為異去氫鉤藤堿、異鉤藤堿、去氫鉤藤堿、鉤藤堿和去氫毛鉤藤堿,18批鉤藤的相似度評(píng)價(jià)表明市場(chǎng)上鉤藤的質(zhì)量差異較大;聚類分析將18批鉤藤飲片樣品聚為3大類;主成分分析用5個(gè)主成分進(jìn)行綜合評(píng)價(jià),其中8批鉤藤飲片的綜合得分大于1;采用一測(cè)多評(píng)法和外標(biāo)法計(jì)算18批鉤藤飲片中5種生物堿的含量,經(jīng)GraphPad差異性分析表明,2種方法無(wú)顯著性差異。結(jié)論 市場(chǎng)上鉤藤質(zhì)量差異較大,建立了檢測(cè)5個(gè)生物堿成分的一測(cè)多評(píng)法和外標(biāo)法,為完善鉤藤質(zhì)量評(píng)價(jià)提供參考。

[Abstract]
Objective To evaluate the quality of Uncaria rhynchophylla collected in Guizhou province based on combinative methods of HPLC fingerprint and multi-components determination and chemical pattern recognition. Methods Tottally 18 batches of U. rhynchophylla were randomly purchased from Guizhou traditional Chinese medicine market. The fingerprint of 18 batches of U. rhynchophylla was established by HPLC. The similarity of 18 batches was analyzed with Similarity Evaluation System for Chromatographic Fingerprint of Chinese Materia (2012). Five alkaloids were identified, and their contents were determined by external standard method and one test and multiple evaluations (QAMS). The hierarchical clustering analysis (HCA) and principal component analysis (PCA) were performed by SPSS 26.0. Results Totally 28 peaks were identified in the fingerprint of U. rhynchophylla and five of the common peaks were identified as isocorynoxeine, isorhynchophylline, corynoxeine, rhyncholphylline, and hirsuteine by reference substance. The fingerprint similarities of the 18 batches indicate that the quality of U. rhynchophylla in the market varied greatly. According to HCA, the samples were divided into three groups. Five principal components were selected for comprehensive evaluation by PCA, and the comprehensive scores of eight batches were greater than one. The contents of five alkaloids in 18 batches were calculated by the methods of external standard method and QAMS. GraphPad was used for difference analysis, and the results showed that there was no significant difference between the two methods. Conclusion This study showed that the quality of U. rhynchophylla differed greatly in the market, we have successfully established the multiple evaluation method and the external standard method for the determination of five alkaloid components, providing a reference for the improvement of the quality evaluation of uncaria hook.
