動物實驗證實厚樸酚及和厚樸酚具有防治急慢性抑郁作用,其主要機制是抗氧化和抗炎:通過直接清除自由基以及提高機體的核因子E2相關(guān)因子-2抗氧化信號通路和抗氧化酶活性間接清除自由基而抑制活性氧生成;通過阻滯磷脂酰肌醇-3蛋白激酶/蛋白激酶B,細胞外信號調(diào)節(jié)激酶/絲裂原活化蛋白激酶和把關(guān)受體/絲裂原活化蛋白激酶信號通路抑制炎性細胞因子表達和小膠質(zhì)細胞、星形細胞的激活,保護腦神經(jīng)。也可增強腦內(nèi)5-羥色胺能神經(jīng)功能,提高腦源性神經(jīng)營養(yǎng)因子表達,促進海馬神經(jīng)再生,產(chǎn)生抗抑郁作用;還可下調(diào)抑郁時的下丘腦-垂體-腎上腺軸功能的亢進,改善抑郁樣行為。

[Abstract]
In animal experiments magnolol and honokiol have the pharmacologic effects in prevention and treatment for acute and chronic depression. Their basic mechanisms are antioxidation and anti-inflammation: by scavenging direct free radial and eliminating indirect free radial through up-regulating Nrf2 anti-oxidative signaling pathway and activity of antioxidases to decrease produce of ROS, and by blocking these signaling pathways of PI3K/Akt, ERK/MAPK and TLR/MAPK to inhibit the expression of inflammatory cytokines, and activation of microgliocyte and astrocyte, and to produce neuroprotective effect in brain. Magnolol and honokiol improve function of 5-hydroxytryptaminergic nerve, and expression of brain-derived neurotrophic factor, and neurogenesis of hippocampus, to produce antidepressant-like effects. Magnolol and honokiol ameliorate depressant-like behaviors by downregulating function of hypothalamus-hypophysis-adrenal axis.
