[摘要]
目的 通過電感耦合等離子體-質(zhì)譜(ICP-MS)法建立同時測定秀麗隱桿線蟲體內(nèi)26種元素含量的方法,同時分析線蟲生長發(fā)育不同階段體內(nèi)元素的含量變化。方法 以65%~68% HNO3作為消解液,采用濕法消解處理線蟲,利用ICP-MS同時測定線蟲中26種元素Al、As、B、Ba、Bi、Ca、Cd、Co、Cr、Cu、Fe、Ga、In、K、Mg、Mn、Mo、Na、Ni、Pb、Sc、Se、Te、V、Zn、Zr。將線蟲同步化后獲得的卵均勻分成5組,分別培養(yǎng)至L1期、L2期、L3期、L4期以及Adult期,應(yīng)用建立的ICP-MS法測定26種元素含量。結(jié)果 26種元素在0~500 μg·L-1線性關(guān)系良好,R2 ≥ 0.999 3;儀器檢出限為0.000 1~0.090 3 μg·L-1;儀器精密度(RSD)為0.4%~1.8%;重復(fù)性(RSD)為0.0%~2.5%;穩(wěn)定性(RSD)為0.8%~4.6%;平均加標(biāo)回收率為80.8%~118.5%,RSD為0.2%~1.5%。L4期線蟲中總體元素含量最高,其次是L2期,L1期線蟲中總體元素含量最低。結(jié)論 通過ICP-MS測定了線蟲生長發(fā)育過程中體內(nèi)元素的含量,該方法快捷靈敏、簡便高效,適用于對線蟲中元素的分布情況進(jìn)行分析,為進(jìn)一步研究單一元素或多種元素對秀麗隱桿線蟲的潛在影響提供參考。

Objective A method was established to simultaneously determine the content of 26 elements in C. elegans by Inductively Coupled Plasma Mass Spectrometry (ICP-MS), and to analyze the content changes of elements in different stages of C. elegans growth and development. Methods The nematodes were treated by wet digestion using 65%-68% HNO3 as the digestion solution, and 26 elements (Al, As, B, Ba, Bi, Ca, Cd, Co, Cr, Cu, Fe, Ga, In, K, Mg, Mn, Mo, Na, Ni, Pb, Sc, Se, Te, V, Zn, and Zr) in the nematodes were determined simultaneously using ICP-MS. The eggs obtained after synchronization of nematodes were evenly divided into five groups and cultured to L1, L2, L3, L4, and Adult stages, respectively. The content of 26 elements was determined using the established ICP-MS method. Results The results showed that the linearity of the 26 elements was good in the range of 0-500 μg·L-1 with R2 ≥ 0.999 3, the instrumental limits of detection were 0.000 1-0.090 3 μg·L-1, the instrumental precision RSD was 0.4%-1.8%, the reproducibility RSD was 0.0%-2.5%, the stability RSD was 0.8%-4.6%, the average spiked recoveries were 80.8%-118.5%, and the RSD was 0.2%-1.5%. The L4 stage nematodes have the highest overall element content, followed by the L2 stage, with the L1 stage nematodes having the lowest overall element content. Conclusion The content of elements in nematodes during growth and development was determined by ICP-MS, which is a fast, sensitive, simple and efficient method suitable for analyzing the distribution of elements in nematodes, and provides a certain theoretical basis for the further study of the potential effects of a single element or a variety of elements on Caenorhabditis elegans.
