[摘要]
目的 研究金釵石斛破壁粉對免疫低下小鼠免疫功能的影響。方法 將180只小鼠隨機(jī)分為6組,每組30只,分別為對照組、模型組、左旋咪唑(陽性對照,ig給予鹽酸左旋咪唑片0.025 g/kg)組及金釵石斛破壁粉低、中、高劑量(0.5、1.0、2.0 g/kg)組。除對照組外,其余5組于給藥前3 d ip 75 mg/kg新配制的環(huán)磷酰胺,每天1次,連續(xù)3 d,制備免疫功能低下小鼠模型。造模完成后每天ig給藥1次,對照組和模型組給予等體積的生理鹽水,連續(xù)14 d。通過計算脾指數(shù)與胸腺指數(shù)、腹腔巨噬細(xì)胞吞噬雞紅細(xì)胞實驗、血清溶血素測定、淋巴細(xì)胞轉(zhuǎn)化試驗及western blotting檢測脾臟中細(xì)胞因子IL-2、TNF-α的蛋白表達(dá),觀察金釵石斛破壁粉對免疫低下小鼠免疫功能的調(diào)節(jié)作用。結(jié)果 與模型組比較,金釵石斛破壁粉1.0、2.0 g/kg顯著升高免疫低下小鼠的脾指數(shù)與胸腺指數(shù),提高巨噬細(xì)胞吞噬功能,升高血清溶血素含量,促進(jìn)淋巴細(xì)胞轉(zhuǎn)化反應(yīng),上調(diào)小鼠脾臟中細(xì)胞因子白細(xì)胞介素-2(IL-2)、腫瘤壞死因子-α(TNF-α)的蛋白表達(dá)(P<0.05、0.01)。結(jié)論 金釵石斛破壁粉顯著增強(qiáng)免疫低下小鼠的免疫功能。

[Abstract]
Objective To observe the effects of Dendrobium nobile Lindl. wall-broken powder (DNLP) on immune function in immunosuppression mice induced by cyclophosphamide. Methods A total 180 mice were randomly divided into six groups, 30 mice in each group, namely control group, model group, levamisole (positive control, ig 0.025 g/kg levamisole hydrochloride tablets) group and low, medium and high dose (0.5, 1.0, 2.0 g/kg) group of DNLP. Except for the control group, the other five groups were ip given 75 mg/kg of cyclophosphamide for three days before administration, once a day for three days to prepare immunodeficient mice model. Mice was ig administered once a day after the completion of the model for 14 consecutive days. The control group and the model group were given saline of the same volume. The effect of DNLP was investigated by spleen index and thymus index, experiment of peritoneal macrophages phagocytizing chicken red blood cells, serum hemolysin test, T lymphocyte proliferation test, and Western blotting to detect IL-2 and TNF-α protein expression of spleen in mice. Results Compared with the model group, 1.0g/kg and 2.0g/kg DNLP significantly increased the spleen index, thymus index, and the content of serum hemolysin, improved the phagocytosis of macrophage, and enhanced the T lymphocytes proliferation, and IL-2 and TNF-α protein expression of spleen in immunosuppression mice (P<0.05 and 0.01). Conclusion DNLP can significantly improve the immune function in immunosuppression mice.
