過氧化物酶體增殖物受體(PPARs)是細(xì)胞能量代謝的主要調(diào)節(jié)因子,PPARs的3種亞型在多種腫瘤細(xì)胞中具有不同的轉(zhuǎn)錄活性與效應(yīng),能量代謝穩(wěn)態(tài)對(duì)細(xì)胞的命運(yùn)至關(guān)重要,關(guān)于腫瘤的能量代謝一直是熱點(diǎn)問題。所有細(xì)胞活動(dòng)都強(qiáng)烈依賴于分解代謝和合成代謝途徑之間的平衡,破壞能量平衡和微環(huán)境,將表現(xiàn)出一系列的代謝改變包括葡萄糖消耗增加,線粒體呼吸的減少,細(xì)胞死亡抗性增強(qiáng),所有這些都是癌癥進(jìn)展的原因。了解癌癥中的代謝過程和闡明PPARs的調(diào)控機(jī)制會(huì)產(chǎn)生新的治療策略。

Peroxisome proliferator-activated receptors (PPARs) are the major regulators of cellular energy metabolism.The three subtypes of PPARs have different transcriptional activities and effects in various tumor cells.And the energy metabolism of tumors has always been a hot spot problem.Energy metabolism homeostasis is critical to cell fate because all cellular activities are strongly dependent on the balance between catabolic and anabolic pathways.Destruction of energy balance and microenvironment will show a cascade of metabolic changes including increased glucose consumption,decreased mitochondrial respiration,and increased cell death resistance,all of which are responsible for cancer progression.Understanding the metabolic processes and elucidating the regulation mechanisms of PPARs will lead to new therapeutic strategies.
