用苯酸-硫酸法進(jìn)行了油松果多糖的含量測定,并考察了苯酚與濃硫酸的用量及反應(yīng)溫度等因素時(shí)顯色反應(yīng)的影響,最大吸收波長490nm,測得多糖在10~70μg/ml的濃度范圍內(nèi)與吸收度呈線性關(guān)系(r=0.9999n=6),平均回收率為99.9%,變異系數(shù)為1.2%,最低檢測濃度為0.5μg/ml。

[Abstract]
Quantitative determination of polysaccharides in Pine cones by phenol-sulfuric acid mehod was studied. and the effects of concentration of phenol-sulfuric acid and reaction temperature on color formation were investigated. It was found that the absorbance is linearly correlated to polysaceharide concentrations between 10~78μg/ml (r=0.9999). The analytical recovery was 99.8%, CV% was 1.2% and the minimal detectable concentration was 5μg/ml.
